Transcriptome analysis of wheat seedlings in response to abscisic acid

ABSTRACT: Screening of an EMS-derived TILLING population based on carbon isotope discrimination and ABA sensitivity during seedling growth identified WS1, a water-saving, drought-tolerant candidate line. We performed transcriptomic analysis to elucidate the regulatory modules underlying the enhanced water-use efficiency and drought tolerance of WS1.

Water-saving and water-spending strategy: The metabolomic and proteomic clarification of the wheat 1 response to drought

Project description:Drought as water deficit in the soil represents the most commonly occurring and the most variable environmental stress factor worldwide. Plants have evolved various strategies to cope with drought stress in relation to their other needs and processes such as the necessity to supply carbon for photosynthetic assimilation in plants with a C3 type of photosynthetic assimilation. In the present study, two wheat cultivars, Baletka and Tobak, representing two contrasting strategies, were studied at physiological, proteomic and metabolomics levels to provide a complex view on their phenotypic responses to drought treatment. Physiological characteristics clearly distinguished Baletka and Tobak and, moreover, in water-saving Baletka both proteomics and metabolomics analyses revealed significant remodelling of cell wall, changes in endocytosis and cell signalling, and especially, changes in synthesis of other defence proteins and LTI 65 kDa protein (also known as RD29B protein), which has been detected in wheat for the first time. An amazing finding was the enhanced accumulation level of oxalate oxidase implying enhanced oxaloxidase activity leading to faster degradation of oxalate with the simultaneous gain of two molecules of CO2, which may represent an alternative source of CO2 for plants with C3 type of photosynthesis suffering under drought stress.

Transcriptomics of water-deficit Stress Responses in TAM Wheat Cultivars

Project description:Water deficit stress between the booting and grain filling stages significantly affect grain yield and quality of hard red winter wheat. Several stress tolerant cultivars with different adaptation mechanisms have been released and are widely cultivated on the Southern Great Plains of the US. However, the physiological, molecular, and genetic basis of adaptation to drought stress for these cultivars remains unknown. Use of transcriptome profiling to identify drought responsive genes in hexaploid wheat is a challenging process given the quantitative nature of drought stress, genome complexity, and the intricacy of interaction effects. If the information generated from functional genomics studies is to be used in molecular breeding programs for cultivar development, it is highly desirable to use cultivars better adapted for the region. In the current study we used two well-adapted, drought-tolerant, high-yielding, cultivated varieties, TAM 111 and TAM 112, which appear to have different adaptation mechanisms, to identify drought stress induced transcripts during heading and early dough stages. A set of 24 Affymetrix GeneChip wheat genome arrays (2 cultivars; 2 water treatments; 2 sampling stages; 3 biological replicates) from plants subjected to water deficit stress under controlled glasshouse conditions. Differentially expressed genes were identified using a ANOVA (p<0.01) controlling false discovery rate (FDR, q<0.01) using Benjamini Hochberg approach.

Tuning water use efficiency and drought tolerance in wheat using ABA receptors

Project description:Water availability is a key determinant of terrestrial plant productivity. Many climate models predict that water stress will increasingly challenge agricultural yields and exacerbate projected food deficits. To ensure food security and increase agricultural efficiency, crop water productivity must be increased. Research over past decades has established that the phytohormone abscisic acid (ABA) is a central regulator of water use and directly regulates stomatal opening and transpiration. In this study, we investigated whether the water productivity of wheat could be improved by increasing its ABA sensitivity. We show that overexpression of a wheat ABA receptor increases wheat ABA sensitivity, which significantly lowers a plant’s lifetime water consumption. Physiological analyses demonstrated that this water-saving trait is a consequence of reduced transpiration and a concomitant increase in photosynthetic activity, which together boost grain production per liter of water and protect productivity during water deficit. Our findings provide a general strategy for increasing water productivity that should be applicable to other crops because of the high conservation of the ABA signaling pathway.

β-diketone accumulation in modern bread wheat varieties

Project description:Cuticular waxes coating leaf surfaces can help tolerate drought events by reducing non-stomatal water loss. Despite their role in drought tolerance, little is known about the cuticular wax responses of Canadian bread wheat varieties. To fill in this gap, RNAseq was performed on the flag leaf of four modern varieties to identify potential markers that could be used for selection of higher accumulation of cuticular waxes. This analysis revealed that the W1 locus is a good candidate for higher accumulation of β-diketones.
