Gene expression profiles of Six1/2-1(RNAi) animals and POU2/3(RNAi) animals

ABSTRACT: The goal of this study was to identify genes that were expressed in the protonephridial system. Because Six1/2(RNAi) and POU2/3(RNAi) animals lose the tubules, flame cells and accessory cells, these animals were expected to show decreased levels of the genes expressed in the nephridial system relative to control RNAi animals.

Project description:The goal of this study was to identify genes that were expressed in the protonephridial system. Because Six1/2(RNAi) and POU2/3(RNAi) animals lose the tubules, flame cells and accessory cells, these animals were expected to show decreased levels of the genes expressed in the nephridial system relative to control RNAi animals. RNA from Six1/2(RNAi) and POU2/3(RNAi) animals was harvested with Trizol at days 8 and 15 after RNAi initiation for the former and days 15 and 21 for the latter. Three biological replicates were used. RNA from the four samples were competetively hybridized on a microarray with RNA from control RNAi animals of the corresponding days.

Project description:The goal of this study was to identify Fir1 downstream genes that were required for local proliferation. Fir1(RNAi) animals displayed reduced local proliferation near the wound,while the proliferation far from the wound was not decreased after Fir1 RNAi. The regenerating pieces following Fir1 RNAi were expected to only show decreased levels of the genes near the wound relative to control RNAi animals.

Expression data from Control and Six1 expressing MCF7 derived xenograft tumors

Project description:Although an important association between lymph node metastasis and poor prognosis in breast cancer was observed decades ago, an active role for the lymphatic system in metastatic dissemination has only recently been examined. We demonstrate that the Six1 homeoprotein promotes peri- and intra-tumoral lymphangiogenesis, lymphatic invasion, and distant metastasis of breast cancer cells. We identify the pro-lymphangiogenic factor, VEGF-C, as required for this process, and demonstrate transcriptional induction as the mechanism of regulation of VEGF-C expression by Six1. Using a different, but complementary animal model, we show that while required, VEGF-C is not sufficient for the pro-metastatic effects of Six1. Verifying the clinical significance of this pro-metastatic Six1-VEGF-C axis, we demonstrate co-expression of Six1 and VEGF-C in human breast cancer. Two Control and two Six1 expressing MCF7 clones were orthotopically injected into NOD/Scid mice forming a total of 3 Control and 3 Six1 tumors that were allowed to grow to 2 cm^3 . The tumors were dissected and total RNA was isolated from each tumor and hybridized to Affymetrix arrays

Differentiation resistant HPV16-immortalized keratinocytes: Six1 transfected vs. Control.

Project description:The overexpression of Six1, a member of the Six family of homeodomain transcription factors, has been found in various human cancers, and is associated with tumor progression and metastasis. We previously determined that the expression of Six1 mRNA increased during in vitro progression of human papillomavirus type 16 (HPV16)-immortalized human keratinocytes (HKc/HPV16) toward a differentiation-resistant (HKc/DR) phenotype. However, if Six1 promotes HPV16-mediated transformation or not remains unknown. HKc/DR were transfected with a Six1 or control vector and RNA isolated from these cells were used in an Agilent two-color gene expression profiling experiment. The goal was to determine the effects of Six1 on global gene expression. Two-condition experiment, Six1 vs. Control HKc/DR cells. Biological replicates: 4 Six1-transfected replicates and 4 control-transfected replicates.
