Ganoderic Acid A alleviates ulcerative colitis by inhibiting the IL-17 signaling pathway via targeting RORA

ABSTRACT: Ulcerative colitis (UC), a chronic inflammatory bowel disease, poses a significant global public health burden, a challenge compounded by the limited availability of effective treatments. Ganoderic acid A (GAA), which is a unique secondary metabolite in Ganoderma lucidum, has demonstrated anti-inflammatory, anti-tumor, antioxidant, and lipid-lowering activities. However, the role and mechanism of GAA in UC are not fully understood. In this study, we aimed to explore the role and involved mechanisms of GAA in UC.

Methyl Gallate Alleviates Acute Ulcerative Colitis by Modulating Gut Microbiota and Inhibiting TLR4/NF-κB Pathway [16S]

Project description:Ulcerative colitis (UC) is a complex immune-mediated inflammatory disease. In recent years, the incidence of UC has increased rapidly, however, its exact etiology and mechanism are still unclear. Based on the definite anti-inflammatory and antibacterial activities of Sanguisorba officinalis L., we studied its monomer methyl gallate (MG). In this study, we employed flow cytometry and detected nitric oxide production, finding MG regulated macrophage polarization and inhibited the expression of proinflammatory cytokines in vitro. MG also exhibited anti-inflammatory activity with ameliorating body weight loss, improving colon length and histological damage in dextran sulfate sodium-induced UC mice. Meanwhile, transcription sequencing and 16S rRNA sequencing analyzed the key signaling pathways and changes in the gut microbiota of MG for UC treatment, proving MG could alleviate inflammation by regulating the TLR4/NF-κB pathway in vivo and in vitro. Additionally, MG altered the diversity and composition of the gut microbiota and changed the abundance of metabolic products. In conclusion, our results are the first to demonstrate MG has obvious therapeutic effects against acute UC, which is related to macrophage polarization, improved intestinal flora dysbiosis and inhibition of TLR4/NF-κB signaling pathway activation, and may be a promising therapeutic agent for UC treatment.

Darutigenol alleviates ulcerative colitis via promoting Akkermansia muciniphila-mediated proline synthesis

Project description:Gut microbiota dysbiosis is associated with the occurrence and development of inflammatory bowel disease (IBD). Darutigenol (DL) is a natural compound with limited oral bioavailability, but has shown promise in the treatment of ulcerative colitis (UC). However, the precise mechanisms underlying its therapeutic effects remain incompletely understood. To identify the critical gut microbial species and microbial metabolites associated with the UC-protective effects of DL, we assessed its microbiota-dependent anti-UC effects through 16S rRNA sequencing, antibiotic treatment, and fecal microbiota transplantation. Metabolomics analysis, transcriptional analysis, and targeted bacteria/metabolites gavage were conducted to explore how DL regulates gut microbiota to reduce the severity of UC. The results showed that DL mainly promoted the synthesis of its metabolite proline by regulating the abundance of Akkermansia muciniphila (A. muciniphila), thereby exerting anti-UC effect. Interestingly, we discovered that DL could promote the growth of A. muciniphila in the gut by targeting IDO1. Additionally, proline supplementation exerts an anti-UC effect by downregulating the IL-17 signaling pathway. Together, our study demonstrated that DL promoted the growth of A. muciniphila by targeting IDO1 and stimulates proline production, thereby alleviating the symptoms of UC. Adenosine-rich extract of Ganoderma lucidum: a safe and effective lipid-lowering substance via regulating expression and modulation of PPAR signaling pathway

Project description:Ganoderma lucidum is a traditional Chinese medicine with a variety of active compounds and possessing adequate lipid-lowering and anti-atherosclerotic effects. However, its main active components and potential mechanisms still remain unclear. Here, we evaluated the anti-hyperlipidemic effect of the adenosine extract from Ganoderma lucidum (AEGL) in high-fat-diet (HFD)-induced hyperlipidemic ApoE-/- mice and explored the underlying biological mechanism by multi-omics analysis. Treatment with AEGL for 8 weeks significantly decreased the serum levels of total cholesterol (TC), triglyceride (TG), and low-density lipoprotein cholesterol (LDL-c) by 45.59%, 41.22%, and 39.02%, respectively, as well as reduced liver TC and TG by 44.15% and 76.23%, compared with the HFD-only group. We also observed significant amelioration of hepatic steatosis without liver and kidney damage after AEGL treatment. Regulating the expression and acetylation/crotonylation of proteins involved in the PPAR signaling pathway may be one of the potential mechanisms involved in the observed lipid-lowering effects of AEGL.

DMLL alleviates ulcerative colitis in mice

Project description:The natural compound DMLL alleviates DSS-induced experimental colitis through modulating gut microbiota and metabolic dysbiosis. The phylum of Bacteroidota and genera of norank_f_Muribaculaceae and Ruminococcus with beneficial potential for UC were the major gut flora regulated by DMLL. In terms of microbiota-derived metabolites, DMLL primarily enriched tryptophan metabolites, secondary bile acids and nicotinamide that possess anti-inflammatory activity and proved protection for intestinal epithelial barrier, subsequently altered the metabolic pathways and improved the impaired host gut homeostasis.
