Project description:Copper (Cu) homeostasis is critical to the health of most organisms, and is thus tightly regulated by several highly conserved processes. Using the unique genomic tools available in yeast, we have expanded current knowledge of these processes by identifying 237 genes important for Cu-dependent growth. 116 of these genes, when deleted, decreased Cu-dependent respiratory growth. The remaining 121 genes, when deleted, produced respiratory growth defects that were specifically rescued by addition of Cu. Among the genes identified by our screen are known regulators of copper homeostasis, genes required to maintain low vacuolar pH, and genes where evidence supporting a functional link with Cu has been heretofore lacking. Many genes identified are not only conserved in man, but also associated with diseases spanning a variety of clinical phenotypes. We demonstrate that the approved drug disulfiram rescues Cu-deficiencies of both environmental and genetic origin, thus underscoring a potential paradigm for treating the broad spectrum of Cu-related disease in man.

Project description:The major objective of this study was to characterise theHapT1 orthotopic hamster pancreatic tumor as a preclinical model of desmoplastic pancreatic ductal adenocarcinoma; and use this model to validate the efficacy of drugs that kills activated pancreatic stellate cells (PSCs)in vitro. Experimentaldesign: Commercially procured HapT1 pancreatic cancer (PCA) cell line was implanted in the pancreas of its syngeneic host, Syrian golden hamster (Mesocricetusauratus). After certain time period the primary and secondary tumors were harvested for histological andimmunophynotypical analysis. PSCs of hamsters were harvested, cultured and characterised. The in-cultured activated rodent PSCs and commercially procured human PSCs were used to check the cytotoxic effect of Disulfiram (DSF) in presence and absence of copper (Cu )in vitro. Finally, theHapT1 orthotopic tumor model was used to check the efficacy of DSF in vivo.

Project description:Some chemotherapeutic agents have been found to enhance the antitumor immunity by inducing immunogenic cell death (ICD). The combination of disulfiram (DSF) and copper (Cu) has demonstrated anti-tumor effects in a range of malignancies including hepatocellular carcinoma (HCC). However, the potential of DSF/Cu as an ICD inducer and whether it could enhance the efficacy of immune checkpoint blockade in HCC remains unknown. Here, we showed that DSF/Cu-treated HCC cells exhibited characteristics of ICD in vitro, such as calreticulin (CRT) exposure, ATP secretion, high mobility group box 1 (HMGB1) release. DSF/Cu treated HCC cells elicited significant immune memory in a vaccination assay. DSF/Cu treatment promoted dendritic cell activation and maturation. The combination of DSF/Cu and CD47 blockade further facilitated DC maturation and subsequently enhanced CD8+ T cell cytotoxicity. Mechanically, DSF/Cu promoted the nuclear accumulation and aggregation of nuclear protein localization protein 4 (NPL4) to inhibit the ubiquitin-proteasome system, thus induced endoplasmic reticulum (ER) stress. Inhibition of NPL4 induced ICD-associated damage-associated molecular patterns. Collectively, our findings demonstrated that DSF/Cu induced ICD-mediated immune activation in HCC and enhanced the efficacy of CD47 blockade.

Project description:Inclusion body myopathy associated with Paget disease of bone and frontotemporal dementia (IBMPFD) is caused by mutations in the Valosin Containing Protein (VCP) gene on chromosome 9p12-13. To elucidate affected signaling transduction axes in IBMPFD, we determined expression profiles using microarray technology in quadriceps muscle from patients and unaffected relatives. Muscle from 10 individuals (7 affected, 3 unaffected first degree relatives) was obtained after informed consent for the muscle biopsy was obtained.

Project description:Disulfiram and niclosamide were identified as drugs that induced depletion of an MLL-AF9-luciferase fusion protein in THP-1 AML cells, in a bioluminescence sreen. MLL-fusion protein depletion was confirmed in AML and ALL cell lines expressing different MLL-fusion proteins. Combination of disulfiram with niclosamide was found to enhance depletion of MLL-fusion proteins. To investigate whether this enhanced depletion resulted in increased suppression of downstream target genes. MLL-AF6 expressing SHI-1 cells were exposed to disulfiram/copper, nciclosamide or combined drugs. SHI-1 cells were treated for 16 hours with 0.3uM disulfiram / 1uM copper, 5uM niclosamide or combined drugs and RNAseq performed on isolated RNA.

Project description:This sstudy was designed to investigate the gene netweork regulated by valosin-containing protein (VCP), an ATPase-associated protein, whic acts as a novel regulator against the cardiac stress of pressure overload. We performed transcriptome analyses to compare cardiac-specific VCP overexpression transgenic (TG) mice and wild-type (WT) mice in a series of pathological models. The left ventricular (LV) tissues were collected from both WT and VCP TG mice at 2w post TAC and at 2w post sham operations. mRNA was extracted and sequenced to generate gene expression profiles. Multiple comparisons were performed between VCP TG and WT mice at two treatment conditions: sham and 2w post TAC. Our studies revealed novel molecular mechanisms by which VCP protects heart against pressure overload-induced cardiac hypertrophy and the transition to the heart failure.

Project description:The NLRP3 inflammasome is dysregulated in autoinflammatory disorders caused by inherited mutations and contributes to the pathogenesis of several chronic inflammatory diseases. In this study, we discovered that disulfiram, a safe FDA-approved drug, specifically inhibits the NLRP3 inflammasome, but not the NLRC4 or AIM2 inflammasomes. Disulfiram suppresses caspase-1 activation, ASC speck formation, and pyroptosis induced by several stimuli that activate NLRP3. Mechanistically, NLRP3 is palmitoylated at cysteine 126, a modification required for its localization to the trans-Golgi network and inflammasome activation which was inhibited by disulfiram. Administration of disulfiram to animals inhibited the NLRP3, but not the NLRC4 inflammasome in vivo. Our study uncovers a mechanism by which disulfiram targets NLRP3 and provides a rationale for using a safe FDA-approved drug for the treatment of NLRP3-associated inflammatory diseases.

Project description:Cholangiocarcinoma (CCA) remains a lethal malignancy with limited therapeutic options. Through genome-wide CRISPR-Cas9 screening, we identified the ATPase valosin-containing protein (VCP) as a critical dependency in CCA. Compound screens revealed that the VCP inhibitor CB-5339 potently suppresses CCA proliferation in a panel of patient derived organoids by inducing cellular senescence. It is known that senescent cells persist and this can contribute to therapy resistance. To address this, we combined CB-5339 with senolytic agents (ABT-263 and conatumumab), which selectively eliminate senescent CCA cells, achieving enhanced tumor suppression in vitro and in vivo. Clinical analysis showed that VCP overexpression in CCA patients correlates with poor prognosis. Our study unveils a “one-two punch” strategy, targeting VCP-mediated senescence followed by senolytic clearance, offering a promising therapeutic approach for CCA.
