Integration of transcriptomics and proteomics uncovers novel targets underlying the protective effects of Nrf2 knockout in HEI-OC1 cells

Project description:Cisplatin, a utilized anticancer drug in clinical practice, induces sensorineural hearing loss (SNHL) in patients. However, the precise mechanism underlying cisplatin-associated ototoxicity remains unknown. HEI-OC1 cells are immortalized cells derived from the organs of Corti mice and nuclear factor erythroid 2-related factor 2 （Nrf2） knockout (KO) significantly enhances cisplatin resistance in these cells. The exploration of transcriptomic data from Nrf2 KO has significant implications for the identification of novel targets to enhance HEI-OC1 cisplatin resistance in Nrf2 KO and for understanding the biological characteristics associated with SNHL. The RNA-seq analysis revealed a significant enrichment of differentially expressed genes (DEGs) in the Nrf2 KO model within key signaling pathways, including the PI3K-Akt, MAPK, as well as Glutathione metabolism signaling pathways. Notably, expression levels of 17 specific genes were confirmed by RT-qPCR (Real-time Quantitative-PCR). The biomarkers identified in this study may be key to understanding the biological mechanism by which Nrf2 KO strongly increases HEI-OC1 cisplatin resistance, and by targeting the PI3K-Akt, MAPK, Glutathione metabolism signaling pathways provide new ideas for the prevention and treatment of cisplatin-induced SNHL.

Transcriptomic in silico screens identify niclosamide for protection against cisplatin- and noise-induced hearing loss in zebrafish and mice through Nrf2 activation

Project description:We report the RNA expression of the House Ear Institute-Organ of Corti (HEI-OC1) cells with and without cisplatin exposure to identify differencialyy expressed genes. The results of RNA-seq analysis showed 2732 downregulated, and 1639 upregulated genes above 1-fold change in cisplatin threated cells compared to non-treated controls.

The cargos and roles of small extracellular vesicles from cochlear explants in cisplatin-induced ototoxicity

Project description:Small extracellular vesicles (sEVs) play an important role in intercellular communication as well as pathological development. Here, to explore the roles of sEVs in the progression of cisplatin-induced ototoxicity, we isolated sEVs from the conditioned culture medium of cisplatin-induced cochlear explants (Cis-sEVs) and the control group (Ctrl-sEVs) and profiled their cargos using label-free LC-MS/MS. Numerous proteins involved in the regulation of the damage response were enriched in the Cis-sEVs. Cltc, Cct2, and Hspa8, which are potentially involved in protein homeostasis and autophagy, are up-regulated in Cis-sEVs compared to Ctrl-sEVs but not in cisplatin-damaged cochlear tissue or in HEI-OC1 cell lysates.

Transcriptome analysis of zebrafish exposed to neomycin and/or tacrine

Project description:Aminoglycosides remain in current use because of the benefits, including the broad-spectrum, inexpensiveness to produce, and a relatively small incidence of allergic reactions. However, aminoglycosides may induce irreversible hearing loss. Recently, tacrine, an acetylcholineesterase inhibitor originally developed to treat Alzheimer’s disease, was found to be protective to neomycin-induced hair cell damage in zebrafish and mouse. However, the protection mechanism remain to be largely unknown. In this study, we revealed that tacrine significantly suppressed the expression of tnf mRNA and ROS production induced by neomycin. Reduction of tnf expression by the antisense morpholino or transcription activator-like effectors nuclease significantly suppressed the hair cell damage but not ROS production induced by neomycin. In zebrafish treated with an antagonist of NMDA receptor MK-801 or a mitochondrial ROS inhibitor acetyl-L-carnitine, ROS production, tnf expression, and hair cell damage induced by neomycin were also significantly reduced. These levels of suppression were not significantly different from those of co-treatment of tacrine and MK-801 or acetyl-L-carnitine. These findings suggest that activation of NMDA receptor followed by ROS production and subsequent tnf expression are involved in neomycin-induced hair cell damage and that tacrine ameliorate neomycin-induced hair cell damage through inhibition of the signaling pathway.

Utricle Neomycin Timecourse

Project description:Much of human hearing loss is caused by loss of auditory hair cell (HC) function. Mammals cannot regenerate these essential mechanoelectrical transducers of sound. However, birds have retained the ability to regenerate HCs from surrounding supporting cells. To better understand hair cell regeneration, we have expression profiled the sensory epithelia from chicken cochleae and utricles. Pure sensory epithelia, consisting of HCs plus supporting cells, were damaged with either neomycin or laser treatment. Changes in gene expression at various points during regeneration were compared to undamaged control cultures on a custom microarray that interrogates the vast majority of transcription factor (TF) genes. These experiments involved multiple biological samples and hundreds of microarray comparisons. Keywords: timecourse
