Project description:Our data demonstrate that lung and bone marrow-derived megakaryocytes (Mks) have distinct immune phenotypes and functions; lung Mks secrete inflammatory cytokines and express molecules that are similar to many tissue resident leukocytes and antigen presenting cells (APCs), and lung Mks process live intact bacteria and present bacteria-derived antigen to CD4+ T cells both in vitro and in vivo. Our in vivo data suggest that lung Mks have important roles in the early activation of T cell responses to a pulmonary pathogen challenge, identifying a novel immune regulatory role for lung Mks.

Project description:Characterize the genes regulated by MKL1/SRF complex in human megakaryocytes (MKs) derived from cord blood or cytapheresis : Using a knock down approach by small interference of MKL1 in MK progenitors, we observed a decrease in the percentage of cells with actin polymerization after adhesion on various substrates, an increase of mean ploidy level and apoptosis. Furthermore, MKL1 inhibition induced a major defect in pro-platelet formation and MKs migration. These results clearly demonstrate that MKL1 is involved in the cytoskeleton organization and maturation of MKs as well as in platelet formation. The goal of gene profiling was to identify new potential MKL1/SRF targets the expression level of which was down regulated after MKL1 inhibition: Human CD34+ cells isolated from cord blood or cytapheresis were transduced by a control lentivirus (designed as SCR; scramble) or by the lentivirus encoding for shRNA of MKL1 (designed as shMKL1) both containing a GFP expressed under the control of PGK promoter. The cells were than grown in serum free medium supplemented by thrombopoietin leading to a generation of megakaryocytes. At day 9 of culture (80% of MKs), the GFP positive cells were sorted, RNAs were extracted and submitted to hybridization as described in annex protocols. Two lists of genes (one for cord blood samples and one for cytapheresis samples) deregulated after the repression of MKL1 were done comparing the intensity of hybridization between SCR and shMKL1 samples. The common list of genes deregulated in MKs from cord blood and cytapheresis after the repression of MKL1 was than generated.

Project description:<p><strong>OBJECTIVE:</strong> To characterize the bile acid (BA) metabolomic profile of umbilical cord blood (UCB) and meconium in healthy newborns.</p><p><strong>METHODS:</strong> Fifteen healthy newborns, which born in the Obstetrics Department of the Affiliated Hospital of Southwest Medical University between July 1 and August 31, 2023, were selected as study subjects. UCB and meconium samples were collected, and BA metabolomics were analyzed using ultra-high performance liquid chromatography-tandem mass spectrometry.</p><p><strong>RESULTS:</strong> UCB predominantly contained primary bile acids (PBAs), whereas meconium was rich in secondary bile acids (SBAs). The PBAs/SBAs ratio in UCB was significantly higher than in meconium [2.64 (2.49, 5.70) vs 0.99 (0.37, 1.58), P < 0.001]. Both UCB and meconium were primarily composed of conjugated BAs. However, the ratio of unconjugated to conjugated BAs was notably higher in UCB than in meconium [0.14 (0.07, 0.18) vs 0.01 (0.01, 0.04), P < 0.001]. In terms of individual metabolite, UCB was dominated by taurochenodeoxycholic acid (TCDCA), taurocholic acid (TCA), and glycochenodeoxycholic acid (GCDCA), while meconium was abundant in TCA, taurohyocholic acid, and TCDCA. Conjugated PBAs in UCB primarily originated from chenodeoxycholic acid (CDCA), whereas those in meconium were mainly derived from cholic acid (CA). The CA/CDCA ratio in conjugated PBAs was significantly lower in UCB than in meconium [0.59 (0.19, 0.75) vs 2.21 (1.34, 3.04), P < 0.001]. SBAs in both UCB and meconium were primarily derived from CDCA, but the CA/CDCA ratio was significantly higher in UCB than in meconium [0.42 (0.21, 0.63), vs 0.03 (0.01, 0.05), P < 0.001]. No PBAs in UCB correlated with their counterparts in meconium, and only three PBAs (TCDCA, GCDCA, and glycochenodeoxycholic acid 3-glucoside in UCB) were correlated with their downstream metabolites in meconium [with hyodesoxycholic acid (HDCA) (r = -0.66, P = 0.01), Tauro-ω-muricholic acid (TωMCA) (r = 0.52, P = 0.048) and ursodeoxycholic acid-7S (r = -0.53, P = 0.04), respectively]. In meconium, most of PBAs were correlated with their downstream metabolites (P all < 0.05): CA was positively correlated with 3-dehydrocholic acid (3-DHCA), TCA was positively correlated with TDCA and 3-DHCA, GCA was positively correlated with 3-DHCA, CDCA was positively correlated with GUDCA, taurolithocholic acid (TLCA), and 7-keto Lithocholic acid (7-KetoLCA) and negatively correlated with isolithocholic acid (isoLCA). TCDCA was positively correlated with THDCA, TUDCA, GUDCA, TLCA, TωMCA, and GHDCA, while GCDCA was positively correlated with TUDCA, GUDCA, TLCA, and GHDCA, and negatively correlated with isoLCA.</p><p><strong>CONCLUSION:</strong> The BA metabolites in UCB and meconium differ significantly, and the downstream BA metabolites in meconium are predominantly correlated with their upstream BAs in meconium, but not those BAs in UCB. Project description:Despite its therapeutic potential and unique immunological properties, the immune composition of umbilical cord blood lacks consistent and comprehensive characterizations. Human umbilical cord blood (UCB) is often discarded after delivery and is difficult to obtain for research purposes. Furthermore, most research on UCB is focused on properties of CD34+ hematopoietic stem cells for transplantation. The Binns Program for Cord Blood Research at Stanford University has the unique advantage of regular collection and isolation of mononuclear cells (MNC) from UCB donors. This study provides a robust characterization of the immune subset compositions of the CD34-negative MNC fraction of UCB (n=50). The study also compares the UCB data to adult peripheral blood (PB) mononuclear cells to identify differences in immune maturity. Using flow cytometry and single-cell RNA sequencing (scRNA-seq), we analyzed UCB and adult PB MNC samples to characterize the cell surface protein and transcriptomic profiles of different immune subsets. Our study findings bring a higher-definition understanding of the unique immunological properties of umbilical cord blood. Study findings reveal a distinct immune profile in UCB, such as a higher average percentage of CD19 B Lymphocytes, CD4 T Cells, CD4 Naive T Cells, CD4 Recent Thymic Emigrants, CD8 Naive T Cells, CD8 Recent Thymic Emigrants, and CD19 Naive B Cells compared to adult PB. Additionally, there were fewer CD19 Memory B Cells in UCB compared to PB. The scRNA-Seq showed concordance in the proportion of immune cell types but captured more differentiated subtypes of cells. Additionally, scRNA-Seq showed unique clustering patterns in UCB, which reflect cell types that converge in adulthood as the immune system matures. These analyses yield the intriguing possibility that the immune heterogeneity of individuals at birth gives way to more stereotyped immune subsets as the immune system is exposed to the external environment and undergoes maturation. Overall, our findings provide a robust characterization of MNC UCB immune subsets and insights into how immune function develops from birth to adulthood.

Project description:Mesenchymal stem cells (MSC) are multipotent cells which can be obtained from several adult and fetal tissues including human umbilical cord units. We have recently shown that umbilical cord tissue (UC) is richer in MSC than umbilical cord blood (UCB) but their origin and characteristics in blood as compared to the cord remains unknown. Here we compared, for the first time, the exonic protein-coding and intronic noncoding RNA (ncRNA) expression profiles of MSC from match-paired UC and UCB samples, harvested from the same donors, processed simultaneously and under the same culture conditions. The patterns of intronic ncRNA expression in MSC from UC and UCB paired units were highly similar, indicative of their common donor origin. The respective exonic protein-coding transcript expression profiles, however, were significantly different. Hierarchical clustering based on protein-coding expression similarities grouped MSC according to their tissue location rather than original donor. Genes related to systems development, osteogenesis and immune system were expressed at higher levels in UCB, whereas genes related to cell adhesion, morphogenesis, secretion, angiogenesis and neurogenesis were more expressed in UC cells. These molecular differences verified in tissue-specific MSC gene expression may reflect functional activities influenced by distinct niches and should be considered when developing clinical protocols involving MSC from different sources. In addition, these findings reinforce our previous suggestion on the importance of banking the whole umbilical cord unit for research or future therapeutic use.
