Transcriptomic analysis in relation to adventitious rooting of different petunia species

ABSTRACT: Excision-induced adventitious rooting of cuttings is a key developmental process for vegetative propagation of many crops, that involves a phytohormone-controlled reprogramming and redifferentiation of specific cells in the stem base. Even though the important role of auxin as inductor of adventitious roots is widely accepted, the endogenous control of auxin action at whole cutting level and the interaction with other hormones are not completely understood. Comparing the genome-sequenced Petunia species P. axillaris and P. inflata, we monitored the transcriptome of phytohormone-related genes in different cutting sections by a phytohormone-targeted microarray. Expression of 17 selected auxin-related genes was quantified in the same RNA samples by RT-qPCR.
