Project description:Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was utilized to analyze adenomyotic and normal myometrial tissues from ten AM patients who underwent hysterectomy with myometrial fibrosis confirmed by Masson staining. This analysis established comprehensive proteomic profiles of adenomyosis patients and revealed widespread alterations in the proteome and metabolome within normal and fibrotic myometrium. Key proteins and signaling pathways linked to myometrial fibrogenesis were identified based on proteomic data.

Project description:Background: Adenomyosis uteri is a chronic gynecological condition frequently coexisting with endometriosis, presenting significant diagnostic challenges due to overlapping symptoms and limitations in imaging techniques. There is a pressing need for reliable, non-invasive biomarkers to enhance diagnostic accuracy and improve patient care. Objective: This pilot study investigated the diagnostic potential of serum and urine microRNA (miRNA) profiles for adenomyosis using machine learning approaches. Methods: Serum and urine samples were collected from 59 patients undergoing surgery for chronic pelvic pain at the Endometriosis Center, RWTH Aachen University Hospital. Among them, 7 had isolated adenomyosis, 34 had histologically confirmed endometriosis, and 18 served as negative controls. miRNA profiling was conducted via next-generation sequencing. A comprehensive feature selection pipeline—including variance filtering, univariate testing, mutual information, and recursive feature elimination—was used to reduce dimensionality. Classification models (Logistic Regression, Random Forest, Support Vector Machine, and Decision Tree) were trained with cross-validation and evaluated using accuracy, precision, recall, F1 score, and ROC-AUC.

Project description:This study aimed to elucidate the role of microRNA miR-92a-3p in the pathogenesis of adenomyosis. We focused on understanding how miR-92a-3p in exosomes derived from ectopic lesions influences the behavior of endometrial cells, DRG neurons, and Human Umbilical Vein Endothelial Cells (HUVECs), and its potential as a non-invasive diagnostic biomarker. Our findings revealed that MiR-92a-3p is significantly upregulated in exosomes derived from ectopic lesions of adenomyosis. This upregulation was associated with enhanced migration and invasion capabilities in eutopic endometrial cells, DRG neurons, and HUVECs. Furthermore, the study demonstrated a significant correlation between the levels of MiR-92a-3p in urinary exosomes and the clinical symptoms of adenomyosis, suggesting its potential as a non-invasive biomarker for the disease. This study elucidates an exosomal signaling process via miR-92a-3p that drives pathological infiltration and angiogenesis to promote adenomyosis progression. Upregulated miR-92a-3p in biofluid exosomes shows promising non-invasive biomarker potential for diagnosis and monitoring of this disease. Our findings unveil novel targets and tools for improved clinical management.

We postulate that dysregulation of genes and pathways in eutopic endometrium may predispose to ectopic implantation. No study, to our knowledge, has examined the global transcriptome of isolated eutopic endometrium from women with clinically significant adenomyosis. Design: Laboratory-based study with full IRB approval and consents. Material and Methods: Endometrial sampling was performed on hysterectomy specimens (proliferative phase) from symptomatic women with pathologically-confirmed diffuse adenomyosis (n=3). Controls (n=5) were normo-ovulatory subjects without adenomyosis. All subjects were free from leiomyoma, endometriosis, and hormonal exposures. Isolated purified total RNA was subjected to microarray analysis using the Gene 1.0 ST Affymetrix platform. Data were analyzed with GeneSpring and Ingenuity Pathway analysis. Validation of several genes was undertaken by QRT-PCR. Results: Comparison of transcriptomes of proliferative endometrium from women with and without adenomyosis revealed 140 up-regulated and 884 down-regulated genes in samples from women with adenomyosis compared to controls. Highly differentially expressed genes include those involved in regulation of apoptopsis, steroid hormone responsiveness, and proteins involved in extracellular matrix remodeling, as well as microRNAs of unknown significance. Affected canonical pathways included eukaryotic initiation factor 2 signaling, oxidative phosphorylation, mitochondrial dysfunction, estrogen receptor signaling, and mTOR signaling. Conclusions: The eutopic endometrium in patients with adenomyosis has fundamental abnormalities that may predispose to invasion and survival beyond the myometrial interface. Project description:Adenomyosis mostly occurs in the females with reproductive age, and the pathogenesis is not clear. If we want to improve the diagnosis and treatment of adenomyosis, we must fully understand the specific molecular mechanism of adenomyosis. The Arraystar Human m6a-mRNA&lncRNA Epitranscriptomic microarray analysis was performed on endometrial specimens of 3 patients in the human adenomyosis group and 3 patients in the normal control group to compare and screen the m6A marker of adenomyosis, providing reference for clinical treatment.

Project description:Adenomyosis is a common gynaecological condition characterised by ectopic endometrial tissue within the myometrium, causing debilitating symptoms with limited treatment options. Despite its prevalence, the pathogenesis of adenomyosis remains poorly understood, hindering the development of targeted therapies for true disease modification rather than symptom management. The objective of this study was to elucidate the spatial transcriptomic profile of adenomyosis lesions with matched eutopic endometrial subregions, with a focus on specific cell types and tissue compartments. The study was designed to identify differentially expressed genes, functionally enriched pathways, and cell-cell communication networks within adenomyosis lesions compared to matched endometrial subregions, and to nominate potential drug targets for future therapeutic development. We show that adenomyosis lesions exhibit a distinct transcriptome intermediate between matched endometrium and myometrium, with enhanced epithelial ciliation and altered biological pathways. We reveal that lesion transcriptome is more like the endometrial basalis than the functionalis, displaying increased olfactory signalling, altered oxidative phosphorylation and ATP synthesis, and an altered immune microenvironment suggestive of chronic inflammation. In silico drug screening identified potential compounds that can reverse the adenomyosis lesion transcriptome. These findings provide insights into the molecular landscape of adenomyosis lesions and pave the way for developing novel, lesion-specific treatments that could potentially spare the eutopic endometrium and offer alternatives to hysterectomy.
