Monocyte-derived pro-angiogenic ocular macrophages localize to neovascular membranes and promote angiogenesis across species

ABSTRACT: We performed a xenium spatial transcriptomics experiment on 6 human donor maculas, 3 with neovascular AMD (consisting of 4 total tissue sections), and 3 without neovascular AMD. Data were loaded into Seurat and cells were classified according to label transfer from previous single-cell RNA sequencing experiments. Macrophage subtypes were classified via label transfer from the M-eYe-verse. Tissue sections were manually traced using the Xenium explorer software to classify cells as residing in the retina, RPE, choroid, choroidal neovascular membrane, or sclera. Differential expression was completed comparing gene expression between choroidal neovascular membrane cells versus non-choroidal neovascular membrane cells
