Project description:While sleep homeostasis is fundamental, its causal contribution on systemic metabolism remains incompletely understood. By decoupling sleep homeostasis from circadian influences, this study identifies a previously unrecognized neural mechanism linking compensatory sleep to whole-body metabolism. Using a minimally confounded sleep homeostasis model, spatial transcriptomics revealed the paraventricular nucleus of the hypothalamus (PVN) as the most transcriptionally active region during rebound sleep. TRAP-tagging and functional manipulations demonstrated that PVN sleep-homeostasis ensemble neurons promote NREM sleep while simultaneously shifting whole-body energy utilization toward lipid oxidation. Activation of these neurons reduced respiratory exchange ratio, downregulated systemic glucose metabolism, and enhanced sympathetic-driven lipolysis in white adipose tissue (WAT), particularly inguinal depot. Stable isotope tracing confirmed suppressed glucose metabolic flux in WAT during recovery sleep. These findings uncover a neural substrate through which sleep homeostasis regulates peripheral energy balance, revealing a metabolic trade-off that may underlie the increased diabetes risk associated with sleep disruption.

Hypothalamic SLC7A14 accounts for aging-reduced lipolysis in white adipose tissue

Project description:<p>The central nervous system has been implicated in the age-induced reduction in adipose tissue lipolysis. SLC7A14 is a lysosomal membrane protein highly expressed in the brain. Herein, we investigated the possible role of hypothalamic SLC7A14 in the age-induced lipolysis reduction. In this study, we demonstrated the expression of SLC7A14 was reduced in proopiomelanocortin (POMC) neurons of aged mice. Overexpression of SLC7A14 in POMC neurons alleviated the age-induced reduction in white adipose tissue (WAT) lipolysis, whereas SLC7A14 deletion mimicked the age-induced lipolysis impairment. Moreover, POMC SLC7A14 regulated WAT lipolysis independently of sympathetic nerves in WAT. Metabolomics analysis revealed that POMC SLC7A14 increased the primary bile acid taurochenodeoxycholic acid (TCDCA) content, which mediated the SLC7A14 knockout- or age-induced WAT lipolysis impairment. Furthermore, SLC7A14-increased TCDCA content is dependent on intestinal apical sodium-dependent bile acid transporter (ASBT), which is regulated by intestinal sympathetic afferent nerves. Finally, SLC7A14 regulated the intestinal sympathetic afferent nerves by inhibiting mTORC1 signaling through inhibiting TSC1 phosphorylation. Collectively, our study suggests the function for central SLC7A14 and an upstream mechanism for the mTORC1 signaling pathway. Moreover, our data provides insights into the brain-gut-adipose tissue crosstalk in age-induced lipolysis impairment.</p>

Depletion of WWP1 increases Adrb3 expression and lipolysis in white adipose tissue of obese mice

Project description:Obesity is defined as abnormal or excessive fat accumulation of body fat and contributes to several metabolic disorders. White adipose tissue (WAT) stores energy in the form of triglycerides and releases energy as free fatty acids and glycerol through a process called lipolysis. People with obesity have impaired catecholamine-stimulated lipolysis, but comprehensive understanding of this lipolysis is still unclear. WW domain-containing E3 ubiquitin ligase 1 (WWP1) is a member of the HECT-type E3 family of ubiquitin ligases and is associated with several diseases. We previously showed that WWP1 expression was increased in WAT of obese mice. In this study, we investigated the function of WWP1 in WAT of obese mice by analyzing the phenotype of Wwp1 knockout (KO) mice fed a high-fat diet. The mRNA levels of beta-3 adrenergic receptor (Adrb3), which were decreased with a high-fat diet, were increased by Wwp1 KO in WAT. Moreover, Wwp1 KO mice showed increased phosphorylated hormone-sensitive lipase levels in WAT and plasma non-esterified fatty acid concentrations. In contrast, noradrenaline and its metabolism were not altered in WAT of obese Wwp1 KO mice. These findings indicate that WWP1, which is increased in adipocytes because of obesity, is a candidate for suppressing the Adrb3 signaling pathway and lipolysis independently of noradrenaline metabolism.
