Project description:Photobiomodulation (PBM) is a non-invasive therapeutic strategy that uses specific light wavelengths to stimulate cellular processes that promote tissue repair, reduce inflammation, and alleviate pain. In this study, we evaluated the efficacy of a high-intensity 980 nm near-infrared (NIR) pulsed laser in a murine model of ovalbumin (OVA)-induced allergic asthma. In OVA-sensitized mice, typical allergic features were noted, including airway wall thickening, significant peribronchial and perivascular inflammatory cell infiltration, and compressed alveolar spaces. NIR PBM treatment markedly attenuated these histological changes. At the molecular level, PBM significantly downregulated type 2 cytokine gene expression, including IL-4, IL-5 and IL-13, in the lung tissue. Flow cytometry analyses further revealed that PBM reduced the infiltration of pulmonary eosinophils and inflammatory monocytes. Transcriptomic profiling coupled with gene set enrichment analysis (GSEA) indicated that PBM suppressed NF-κB-mediated inflammatory signaling while modulating Th2-related responses. Collectively, our findings demonstrate that high-intensity 980 nm NIR PBM effectively mitigates key features of allergic asthma in a murine model, supporting its potential as a non-pharmacological, non-invasive adjunct therapy for asthma management.

Project description:Aim: To identify the genes and non-coding RNAs (ncRNAs) involved in the neuroprotective actions of a dietary anti-oxidant (saffron) and of photobiomodulation. Methods: We used a previously published assay of photoreceptor damage, in which albino Sprague Dawley rats raised in dim cyclic illumination (12h 5 lux, 12h darkness) are challenged by 24h exposure to bright (1,000 lux) light. Experimental groups were protected against light damage by pretreatment with dietary saffron (1mg/kg/day for 21d) or photobiomodulation (10 J/cm2 at the eye, daily for 5d). RNA from 1 eye of each of 4 animals in each of the 6 experimental groups (control, light damage (LD), saffron, photobiomodulation (PBM), saffronLD, and PBMLD) was hybridized to Affymetrix rat genome ST arrays. Quantitative real-time PCR analysis of 14 selected genes was used to validate microarray results. Results: LD caused the regulation of 175 entities (genes and ncRNAs) beyond criterion levels (P < 0.05 in comparisons with controls, fold-change >2). PBM pretreatment reduced the expression of 126 of these 175 LD-regulated entities below criterion; saffron pretreatment reduced the expression of 53 entities (50 in common with PBM). In addition, PBM pretreatment regulated the expression of 67 entities not regulated by LD, while saffron pretreatment regulated 122 entities not regulated by LD (48 in common with PBM). PBM and saffron, given without LD, regulated genes and ncRNAs beyond criterion levels, but in lesser numbers than during their protective action. A high proportion of the entities regulated by LD (>90%) were known genes; by contrast, ncRNAs where prominent among the entities regulated by PBM and saffron in their neuroprotective roles (73% and 62% respectively). Conclusions: Given alone, saffron and (more prominently) PBM both regulated significant numbers of genes and ncRNAs. Given prior to retinal exposure to damaging light, thus while exerting their neuroprotective action, they regulated much larger numbers of entities, among which ncRNAs were prominent. Further, the downregulation of known genes and of ncRNAs was prominent in the protective actions of both neuroprotectants. These comparisons provide an overview of gene expression induced by two neuroprotectants and provide a basis for more focused study of their mechanisms. The were 3 biological repliactes of each of the following groups: Control, Saffron pretreated, Photobiomodulation pretreated, Light Damage, Saffron Light Damage and Photobiomodulation Light Damage. 18 chips in total were performed.

Project description:Neuroinflammation plays an important role in the occurrence and development of neurological diseases. In addition to microglia, the role of astrocytes in neuroinflammation has gradually attracted attention. Photobiomodulation (PBM), as a non-invasive, safe and effective treatment, has been shown to alleviate neuroinflammation and show potential in improving astrocyte activation. In this study, by analyzing the effects at different times, the spatiotemporal regulation and molecular mechanism of PBM on astrocytes were deeply explored. The results showed that PBM significantly attenuated LPS-induced upregulation of inflammatory factors and mitochondrial dysfunction in astrocytes under LPS stimulation for 4 hours and 24 hours. RNA-seq analysis showed that the JAK-STAT pathway played an important role in the early stage of astrocyte inflammatory response. PBM can promote the expression of Socs3 by activating Stat5a translocation to the nucleus, further inhibit the overactivation of the JAK-STAT inflammatory signaling pathway, and thus alleviate astrocyte inflammation. Taken together, this study provides new insights into the molecular mechanism of the potential application of PBM in the treatment of neuroinflammation.

Project description:The purpose of the experiment was to determine whole transcriptome changes after spinal cord injury (SCI) to understand how photobiomodulation promotes neuroprotection and functional recovery. SCI was performed at the thoracic (T) level T8, by crushing the dorsal columns using a calibrated watchmaker's forceps. Animals were then treated with photobiomodulation (PBM; 660nm wavelength) or sham control light, within 15 minutes of the injury. PBM or sham-treatment was treatment directed at the lesion site for 1 minute duration every 24hr for the first 3 days. Animals were then killed, tissues harvested and the RNA was extracted using RNEasy lipid tissue mini kit (Qiagen) according to the manufacture's instructions. Whole genome sequencing was outsourced to Qiagen RNA Sequencing Services (Germany).Library preparation was performed using the QIAseq Stranded Total RNA Library Kit with QIAseq FastSelect rRNA and globin depletion. QIAseq FastSelect rRNA HMR was used to reduce the amount of unwanted RNA species. After first and second strand synthesis, the cDNA was end-repaired and 3’ adenylated. Sequencing adapters were ligated to the overhangs. Adapted molecules were enriched using 16 cycles of PCR and purified by a bead-based cleanup. Library preparation was quality controlled using capillary electrophoresis (High Sensitivity Tape D1000) and high-quality libraries were pooled based on equimolar concentrations. The library pool(s) were quantified using qPCR and optimal concentration of the library pool used to generate the clusters on the surface of a flowcell before sequencing on a NovaSeq (Illumina Inc., Madison, USA) instrument (2x75, 2x10) according to the manufacturer instructions (Illumina Inc.). Raw data was de-multiplexed and FASTQ files for each sample were generated using the bcl2fastq software v2.20.0.422 (Illumina inc.).

Project description:Alzheimer’s disease (AD) is the leading cause of dementia worldwide, and despite the passage of over a century since its discovery, we have yet to discover a cure. Hippocampal synaptic dysfunction driven by toxic amyloid-β oligomers (AβO) is an early event in the progression of neurodegeneration and cognitive decline in AD. Non-invasive therapies such as photobiomodulation (PBM) are receiving increased attention in AD research due to the accessibility and relative safety compared to novel drug trials, and have been shown to induce synaptic resilience in animal models of AD. Nano-pulsed laser therapy (NPLT) is a type of PBM therapy using pulsed 808nm near-infrared laser light and optoacoustically generated ultrasound waves to stimulate deeper brain structures than would be accessible by traditional PBM therapy. Here we use NPLT to stimulate adult hippocampal neural stem cells (NSC) and differentiate them in vitro to assess resilience of the mature neurons against AβO toxicity. We found that neurons differentiated from NPLT-treated NSC are resilient against AβO binding and mitochondrial toxicity, and show increased expression of genes associated with autophagy and proteostasis.

Project description:Photobiomodulation (PBM) with blue light induces a biphasic dose response curve in proliferation of immortalized human keratinocytes (HaCaT), with a maximum proliferative effect reached with 7.5min (10.35J/cm2). The aim of this study was to test the photobiomodulatory effect of 10.35J/cm2 blue light irradiation on cell proliferation, ROS production and gene expression at different time points after irradiation of HaCaT cells in vitro and to compare the results with the anti-proliferative phase of PBM using blue light with 41.4J/cm2. For 10.35J/cm2 cell proliferation was increased for all measured harvesting time points up to at least 24h after irradiation. ROS concentration was increased 30min after irradiation. However, already 1h after irradiation, cells were able to decrease ROS and stabilize the concentration to the basal level. The sudden increase in ROS was even higher when compared to 41.4J/cm2. However, gene expression analysis did not indicate any sign of apoptosis and even indicated an activation of cell survival mechanisms. Furthermore, comparable to 41.4J/cm2, aryl hydrocarbon receptor (AHR) “battery genes” showed a deregulation after blue light irradiation strengthening the hypothesis of AHR as possible target for blue light irradiation.
