Endothelial NAD-dependent return to quiescence is required for angiogenesis

ABSTRACT: The process of angiogenesis requires endothelial cells (ECs) to toggle between states of quiescence versus high replication, migration, and tissue invasion. Little is known regarding how ECs return to quiescence during this process. This project aims to understand the role of NAD in endothelial cell proliferation and the transition of from proliferation to quiescence.

Quiescent endothelial cells upregulate fatty acid β-oxidation for vasculoprotection via redox homeostasis

Project description:Little is known about metabolic changes accompanying endothelial cell (EC) quiescence. Nonetheless, when dysfunctional, quiescent ECs (QECs) contribute to multiple cardiovascular diseases. ECs need fatty acid β-oxidation (FAO) for proliferation. Surprisingly, we now report that QECs are not hypo-metabolic, but upregulate FAO >3-fold higher than proliferating ECs (PECs), not to support biomass or energy production, but to sustain the TCA cycle for redox homeostasis through NADPH production. Hence, inhibition of FAO-controlling CPT1A promotes EC dysfunction (anti-fibrinolysis, leukocyte infiltration, barrier disruption) by increasing oxidative stress in CPT1AΔEC mice with endothelial CPT1A loss. Mechanistically, Notch1 orchestrates the use of FAO for redox balance in QECs. Supplementation of acetate (metabolized to acetyl-CoA) induces vasculoprotection against oxidative stress and EC dysfunction in CPT1AΔEC mice, possibly creating therapeutic opportunities. Thus, ECs use FAO for vasculoprotection against their high oxygen (oxidative stress-prone) milieu, and for different metabolic purposes dependent on their proliferation versus quiescence status.

Tubastatin A maintains skeletal muscle stem cell (MuSC) quiescence

Project description:We show that Tubastatin A (TubA) preserves MuSC quiescence and stem cell potency ex vivo, by inhibiting HDAC6 and, consequently, primary cilium resorption. Treatment with TubA improves MuSC engraftment potential and induces a return to quiescence in cycling MuSCs, revealing a potentially valuable approach to enhancing the therapeutic potential of MuSCs. To examine the state of quiescence preserved by TubA at the transcriptome level, we performed RNA-Seq and we found that TubA-treated MuSCs exhibit a quiescent transcriptome. The molecular mechanisms involved in the maintenance of quiescence by TubA were ribosome- and oxidative phosphorylation-related genes as well as low expression levels of cell cycle genes and Hh signaling genes.

Endothelial sensing of AHR ligands regulates intestinal barrier immunity [AHR KO RNA-seq]

Project description:The blood and lymphatic vasculature is lined by functionally specialised endothelial cells (ECs). In the intestine, ECs act as an essential physical barrier, controlling nutrient transport, facilitating tissue immunosurveillance, and coordinating angiogenesis and lymphangiogenesis to ensure appropriate tissue perfusion and drainage. Conversely, endothelial maladaptation can lead to pathological angiogenesis and the perpetuation of inflammation in chronic inflammatory diseases. However, whether enteric ECs actively engage in the regulation of intestinal homeostasis and pathology through integration of environmental cues is currently unknown. Here, we show that the aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor, acts as critical node for EC-sensing of dietary and microbial metabolites. We first established a comprehensive single-cell endothelial atlas of the mouse small intestine uncovering the cellular complexity and functional heterogeneity of blood and lymphatic ECs, identifying transcriptional networks and putative biological roles across endothelial subtypes. Analyses of AHR mediated responses at single-cell resolution identified tissue-protective transcriptional signatures and regulatory networks promoting quiescence and vasculoprotection. Endothelial AHR-deficiency in mice resulted in cellular activation, proliferation, and initiation of angiogenic pathways disrupting tissue homeostasis. In human ECs, AHR signalling promoted quiescence through cell cycle arrest and tempered endothelial inflammatory responses. Together, our data provide a comprehensive dissection of the impact of environmental sensing across the spectrum of enteric endothelia, demonstrating that endothelial AHR signalling serves to promote homeostasis and prevent aberrant inflammatory responses at the intestinal barrier.
