Effect of IL6 depletion on gene expression in Arid1a/Pik3ca Co-mutant mouse Ovarian Tumor Cell Lines

ABSTRACT: Ovarian clear cell carcinoma is a refractory gynecological malignancy resistant to chemotherapy, with 50% exhibiting Arid1a/Pik3ca co-mutations and known to be IL6-high-producing tumors. Despite possessing an immunosuppressive tumor microenvironment, some cases have been reported to respond remarkably well to immune checkpoint inhibitors, though the molecular mechanisms underlying these responses remain unclear. Therefore, to confirm the role of IL6 in tumor immunity, we established cell lines from mouse model tumors with Arid1a/Pik3ca co-mutations that develop ovarian tumors. We created IL6 knockdown cell lines and compared gene expression.
