Project description:Strigolactones (SLs) are carotenoid-derived plant hormones that control shoot branching and communications between host plants and symbiotic fungi or root parasitic plants. Extensive studies have identified the key components participating in SL biosynthesis and signaling, whereas the catabolism or deactivation of endogenous SLs in planta remains largely unknown. Here we report that the Arabidopsis carboxylesterase 15 (AtCXE15) and its orthologues function as efficient hydrolases of SLs. We show that overexpression of AtCXE15 promotes shoot branching by dampening SL-inhibited axillary bud outgrowth. We further demonstrate that AtCXE15 could bind and efficiently hydrolyze SLs both in vitro and in planta. We also provide evidence that AtCXE15 is capable to catalyze hydrolysis of diverse SL analogues and that such CXE15-dependent catabolism of SLs is evolutionarily conserved in seed plants. These results disclose a catalytic mechanism underlying homeostatic regulation of SLs in plants, which also provides a rational approach to spatial-temporally manipulate the endogenous SLs and thus architecture of crops and ornamental plants.

Project description:Strigolactones (SLs) are plant hormones that regulate diverse developmental processes and environmental responses in plants. It has been discovered that SLs play an important role in regulating plant immune resistance to pathogens, but there are currently no reports on their role in the interaction between Nicotiana benthamiana and Tobacco mosaic virus (TMV). In this study, the exogenous application of SLs weakened the resistance of N. benthamiana to TMV, promoting TMV infection, whereas the exogenous application of Tis108, an SL inhibitor, resulted in the opposite effect. Virus-induced gene silencing (VIGS) inhibition of two key SL synthesis enzyme genes, NtCCD7 and NtCCD8, enhanced the resistance of N. benthamiana to TMV. Additionally, we conducted a screening of N. benthamiana related to TMV infection. TMV-infected plants treated with SLs were compared to the control by using RNA-seq. KEGG enrichment analysis and weighted gene co-expression network analysis (WGCNA) of differentially expressed genes (DEGs) suggested that plant hormone signaling transduction may play a significant role in the SL-TMV-N. benthamiana interactions. This study reveals new functions of SLs in regulating plant immunity and provides a reference for controlling TMV diseases in production.

Project description:In maize, nitrate regulates root development thanks to the coordinated action of many players. In this study, the involvement of SLs and auxin as putative downstream components of the nitrate regulation of lateral root development was investigated. To this aim, the endogenous SL content of maize root in response to nitrate availability was assessed by means of LC-MS/MS and measurements of lateral root density in the presence of analogues or inhibitors of auxin and strigolactones were performed. Furthermore, un untargeted RNA-seq based approach was used to better characterize the participation of auxin and strigolacotones to the transcriptional signature of maize root response to nitrate. Our results suggested that N deprivation toughly induces zealactone and carlactonoic acid biosynthesis in maize root, to a higher extent if compared to P-deprived roots. Moreover, data on lateral root density led to hypothesise the existence of both auxin-dependent and auxin-independent effects of nitrate on LR development. In addition, the inhibition of SL biosynthesis seems to participate to the auxin-dependent induction of LR, but the involvement of further downstream unknown components cannot be ruled out.

Project description:The aim of this study was to investigate the role of strigolactones (SLs) in controlling tillering and their involvement as a signal for nitrogen status. RNA-seq analysis was performed on 18-day-old hydroponically grown Tad17 triple knock-out TILLING mutant plants (SL-deficient mutant) and WT-segregant plants, 8 days after introducing them to nitrogen limiting conditions. The experiment consisted of two different treatments: High Nitrogen (10 mM N) and Low Nitrogen (0.1 mM N). Each treatment included six biological replicates. Total RNA was extracted from pooled samples of four basal node sections per biological replicate. In this study, the basal node was defined as the 0.5 cm section from the base of the main shoot, comprising the apical meristem, lateral buds, leaf meristems, etc.

DWARF53 (D53) is a key repressor in the SL signaling. Nevertheless, its involvement and regulatory mechanisms in cotton fiber development—a representative single-cell model, has remained elusive. In this study, we unveil GhD53 represses the transcription of SL-inducible genes GhFAD3-2 and GhFAD3-4, which are involved in linolenic acid biosynthesis—a stimulant of cell elongation, thus inhibiting cotton fiber elongation. We demonstrate that GhD53's inhibitory influence on cotton fiber elongation is relieved in the presence of SLs, as GhD53 undergoes degradation. This alleviation of GhD53 repression triggers the activation of GhFAD3 expression, promoting fiber cell elongation. Remarkably, GhD53 serves as a dual-function transcription repressor, impeding SLs’ role in fiber elongation. Firstly, as a transcription factor, GhD53 directly binds to GhFAD3 promoters, suppressing their transcription. Secondly, GhD53 physically interacts with GhSLRF, a bHLH-type transcription factor, preventing it from binding to GhFAD3 promoters and repressing its transcription activation activity. GhD53 emerges as a pivotal component in the regulatory network governing SL signaling transduction. Overall, our findings provide novel insights into D53’s role in SL signaling and delineate a comprehensive framework for transmitting SL signals to modulate plant cell elongation behavior.

Project description:Strigolactones (SLs) are plant hormones involved in plant growth, stress response and plant-organism interactions. DWARF14 (D14) is a α/β hydrolase involved in the perception of SLs in plants. Zaxinone has been reported as a plant endogenous apocarotenoid that modulates growth and biotic interactions, however, the molecular basis of its perception remains elusive. Here we identified a new D14 ligand, the apocarotenoid zaxinone. We showed that hundreds of genes, and specifically CAROTENOID CLEAVAGE DEOXYGENASE7 (CCD7) and CCD8, require At/OsD14, and at lower extent MORE AXILLARY BRANCHING2 (MAX2)/D3, for their transcriptional induction. We find that zaxinone and D14 physically interact in vitro with lower affinity than GR24; the resolution of the AtD14-zaxinone complex, by X-ray crystallography, provided additional evidence that zaxinone antagonizes SL by binding D14. Zaxinone treatments in d14 mutants revealed that At/OsD14 are essential to trigger the induction of CCD7 and CCD8 genes, SL content (roots and exudates) and Striga seed germination validating their interaction in planta. Moreover, OsD14 is required to trigger the zaxinone growth-promoting effect in roots. These findings reveal a perception mechanism for the hormone candidate zaxinone through the SL receptor D14 to regulate transcription of SL genes, SL content and growth in higher plants.
