Gene expression profiling in HEK293 cells overexpressed lactoferrin gene that has the signal sequence (sLF) or not (ΔLF)

ABSTRACT: Human lactoferrin (LF) is a multifunctional protein involved in immunomodulation, cell growth, and differentiation. In addition to the secreted form (sLF), an alternatively spliced form (ΔLF) that lacks the signal sequence and downregulated in cancer was found. This study was carried out to identify and compare signaling networks provoked by the two LF isoforms. To do this, the two forms were overexpressed in HEK293 cells using the flp-in/tet-on system and genome-wide expression analysis of 18,367 genes was conducted.

Global transcriptome analysis of paracancerous lung tissues from the B16 metastasis mice model under the influence of lactoferrin knockout

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are using transcriptome profiling (RNA-seq) to evaluate the effects of lactoferrin deficiency on the global transcriptome of the lung microenvironment of the B16 metastasis mice model. Methods: 2×10^5 B16-F10 cells were injected into WT and lactoferrin kncoutout (Lf-/-) mice (each group has 10 eight-weeks-old male mice) through tail vein. At 3 weeks post injection,the paracancerous lung tissue were isolated (not including any metastatic colonies). RNAs were extracted by Trizol and sequenced by Solexa high-throughput sequencing service (Oebiotech, Shanghai, China). Data were extracted and normalized according to the manufacturer’s standard protocol.Each group has two mice paracancerous lung tissues be tested. Results: There are significant gene expression profile changes between WT and Lf-/- tumor-bearing mice. Conclusions: Our study describes the global transciptome changes of paracancerous lung tissues from the B16 metastasis mice model under the influence of lactoferrin kncokout.

Metabolomic profiling reveals the effects of early-life lactoferrin intervention on protein synthesis, energy production and antioxidative capacity in the liver of suckling piglets

Project description:<p>The objective of this study was to reveal the effects of early life lactoferrin intervention on the liver metabolism in the suckling piglets. Sixty newborn piglets with an average initial body weight (BW) of 1.51 ± 0.05 kg were assigned to the control (CON) group and the lactoferrin (LF) group. Piglets in the LF group were orally administered LF solution (0.5 g/kg BW per day) from 1 to 7 days of age, and the piglets in the CON group were orally administrated with the same dose of physiological saline. Plasma, jejunum and liver samples were collected on day 8 and 21. The LF piglets had a decreased level of plasma urea nitrogen on day 8 and an increased level of plasma albumin on day 21. Pathway analysis of metabolomic profiles showed that LF treatment affected amino acid metabolism in the liver of piglets. Additionally, LF treatment upregulated the gene expression of proteolytic enzyme and amino acid transporters (APA, aminopeptidases A; APN, aminopeptidases N; EAAC 1, excitatory amino acid carrier 1; Pept 1, peptide transporter 1) in the jejunum, and enhanced phosphorylation levels of mTOR and p70S6K in the liver. Moreover, LF treatment upregulated the expression of β oxidation related gene (CPT 1, carnitine palmitoyl transferase-1) and affected tricarboxylic acid (TCA) cycle in the liver on day 21. Furthermore, LF piglets had a lower level of MDA and a higher level of GSH and GSH-px in the liver mitochondria. Overall, the early life LF intervention affected protein synthesis, energy production and antioxidative capacity in the liver of neonatal piglets.</p>

A 17 kDa fragment of lactoferrin associates with the termination of inflammation and peptides within promote resolution

Project description:During the resolution of inflammation macrophages engulf apoptotic polymorphonuclear cells (PMN) and can accumulate large numbers of their corpses. Here we report that resolution-phase-macrophages acquire the neutrophil-derived glycoprotein lactoferrin (Lf) in vivo and ex vivo and process it to short fragments. During the onset and resolving phases of inflammation in murine peritonitis and bovine mastitis Lf fragments of 15 and 17 kDa occurred in various body fluids, and the murine fragmentation and release were mediated by macrophages. The 17 kDa fragment contained two bioactive tripeptides, FKD and FKE that promoted human macrophage reprogramming to a pro-resolving phenotype. At low concentrations (1-10 M) this was reflected by inhibition of LPS-induced TNFand IL-6 secretion and increased IL-10 levels. Both peptides inhibited ERK and cJun activation following macrophage exposure to LPS. In addition, FKD, and to a lesser extent FKE, promoted neutrophil-mediated resolution at high concentrations (30-100 M) by enhancing the formation of cytokine-scavenging aggregated NETs at low cellular density.
