ABSTRACT: Potentially carcinogenic mutations are widespread throughout the skin, and it is unknown why only some sites develop into a malignancy while most others continue to function normally. Using 10x Genomics Chomium Flex scRNA-seq, we explored differences in skin with similar levels of photodamage adjacent to and proximal to (5cm away from) a melanoma excision scar, and distant photoprotected skin.

α-Arbutin Prevents UVA-Induced Skin Photodamage via Alleviating DNA Damage and Collagen Degradation in NIH-3T3 cells

Project description:Ultraviolet radiation (UV) causes certain side effects to the skin, and their accumulation to a certain extent can lead to accelerated aging of the skin. Recent studies suggests that α-arbutin may be useful in various disorders such as hyperpigmentation disorders, wound healing, and antioxidant activity. However, the role of α-arbutin in skin photodamage is unclear. In this study, under UVA-induced photodamage conditions, α-arbutin treated mouse skin fibroblasts (NIH-3T3) can repair DNA damage and resist apoptosis by reducing the production of reactive oxygen species (ROS) and increasing the phosphorylation of glycogen synthase kinase 3 beta (GSK3β) to orchestra AKT/GSK3β pathway. Meanwhile, α-arbutin can also regulate collagen metabolism and facilitate the replenishment of collagen by targeting the phosphorylation of SMAD3 to mediate the TGFβ/SMAD pathway in NIH-3T3. In conclusion, we found that α-arbutin can mitigate the detrimental effects of skin photodamage induced by UVA irradiation, and provides a theoretical basis for the use of α-arbutin in the treatment of skin photodamage.

Small RNA Profiling of Lesional Psoriatic and Adjacent Normal Skin Tissues

Project description:Purpose: To identify the differentially expressed miRNA in lesional skin of psoriasis patients compared to adjacent normal tissue by Next Generation Sequencing. Methods: Small RNA isolated from 24 paired lesional and adjacent normal tissue sample and Next Generation Sequencing was performed. The sequences reads were trimmed of adapter sequences and quality trimmed using CutAdapt, followed by alignment to the human genome via 'Novoalign'. The aligned files were sorted based on chromosomal location and HTSeq-count was used to obtain the raw read counts for all known miRNAs. Differnetial expression analysis was performed with the raw reads using R-package 'edgeR'. MiRNA expression was quantified in tissue and serum by qRT-PCR method using SYBR-Green assay. Correlation analysis was done between miRNA expression in individual samples with their respective PASI scores to identify the miRNA associated with disease severity. Results: We identified 75 differentially expressed miRNA with at least 2-fold deregulation that were statistically significant. qRT-PCR validation showed approx 90% concordance with NGS data. Fold change of 4 miRNAs - miR-30b-5p, miR-7-1-3p, miR-33a-3p and miR-4742-3p were found to be asssociated with disease severity in psoriasis skin. Another miRNA let-7d-5p expression was found to be associated with disease severity in serum of HLA-Cw6 positive patients. Conclusions: Our study identified several unqiue deregulated miRNAs in psoriatric skin, which had not been previously reported. We also identified miRNA whose deregulated expression in psoriasis is associated with disease severity and the biological pathways through which they regulate disease pathogenesis.

Whole Transcriptome Profiling of Lesional Psoriatic and Adjacent Normal Skin Tissues

Project description:Purpose: To identify the differentially expressed genes in lesional skin of psoriasis patients compared to adjacent normal skin tissue by Next Generation Sequencing. Methods: Total RNA was isolated from 24 paired lesional and adjacent normal skin tissue samples and Next Generation Sequencing was performed. The sequences reads were trimmed of adapter sequences and quality trimmed using CutAdapt, followed by alignment to the human genome (hg19/GRCh37) by 'STAR (v2.7.10a)'. The aligned files were sorted based on chromosomal location. Trancript assembly was performed using StringTie (v1.3.3b) and python script prepDE.py was used to obtain the raw read counts for all annotated genes. Differnetial expression analysis was performed with the raw reads using R-package 'edgeR'. Gene ontology with the deregulated genes was performed uign DAVID GO. The differentially expressed genes were integrated with differentially expressed miRNAs (GSE183547) and differentially methylated CpGs (GSE115797) to identify the genes regulated by miRNA and DNA metylation respectively. Results: We identified 1,541 differentially expressed protein-coding genes with at least 2-fold deregulation that were statistically significant (FDR ≤ 0.05). The deregulated genes were found to be associated with biological processes including inflammatory response, cytokine mediated signaling, keratinization, neutrophil chemotaxis and defense response to virus among the top enriched processes. Inetgrative analysis with small RNA-seq and DNA methylation profiling using Illumina Infinum 450K BeadArray identified 487 and 321 genes to be regulated by miRNA and DNA methylation respectively. Conclusions: Our multi-omics integrative analysis identfied the biological processes regulated exclusively as well as combinatorially by miRNAs and DNA methylation in the disease pathogenesis of psoriasis.

Protective Effect of Ginsenoside Re against UVB-Induced Photodamage in Rats

Project description:Background: With the growing prominence of skin photodamage caused by ultraviolet (UV) radiation, the development of efficient and safe natural photoprotectants has become a major research focus. Ginsenoside Re, a rare active component of Panax ginseng, has attracted much attention due to its significant antioxidant and anti-inflammatory activities, but its systemic role and mechanism in protecting against photodamage remain unclear.  Objective: To exploring the mechanism behind the therapeutic effect of Re on UVB-Induced skin photodamage in a rat model Methods: In this study, a UVB-induced rat photodamage model was established to evaluate the protective effect of ginsenoside Re through histopathological staining, biochemical index detection, etc. and combined with transcriptome and metabolomics analysis to screen differentially expressed genes and metabolites, thereby revealing metabolic pathways and gene functions associated with the photodamage phenotype. Key targets were further verified through multi-omics integration and Western blotting.Results: The results showed that ginsenoside Re could effectively alleviate UVB-induced pathological injury of skin tissue, reduce the level of oxidative stress and the release of inflammatory factors. In addition, it could effectively reverse the abnormal expression of 265 differential genes and 30 metabolites. Through multi-omics association analysis and WB verification, the glutathione metabolism pathway was identified as a key pathway mediating the protective effects of ginsenoside Re against skin photodamage.

Synergistic Effects of Retinol and Retinyl Palmitate in Alleviating UVB-Induced DNA Damage and Promoting the Homologous Recombination Repair in Keratinocytes

Project description:Ultraviolet B (UVB) rays are a type of ultraviolet radiation emitted by the sun, primarily responsible for skin photodamage. These rays mainly affect the epidermis, leading to direct damage to DNA and contributing to skin cancer development. Retinol and its derivatives are effective in combating skin aging and photodamage, but they often cause skin intolerance, limiting their use despite their potent effects. Therefore, investigating optimal compositions of retinoids is essential to enhance their efficacy against photodamage. In this study, we aimed to investigate the synergistic effects of retinol (ROL) and retinyl palmitate (RPalm) in alleviating UVB-induced DNA damage in human keratinocytes (HaCaT). Applying the ROL+RPalm combination after UVB exposure in HaCaT cells significantly reduced inflammation and apoptosis while promoting collagen synthesis compared to individual treatments with ROL or RPalm. Through bulk mRNA sequencing, we identified that the biological function of the ROL+RPalm synergy is primarily directed toward mediating DNA damage repair. Using comet assays, Western blotting, immunofluorescence, and flow cytometry, we demonstrated the anti-DNA damage effects of ROL+RPalm in HaCaT cells and reconstructed human epidermis. We further elucidated that the molecular mechanism underlying the ROL+RPalm combination involves the activation of RARβ, which subsequently triggers the ATM-CHK2-p53 signaling pathway and increases the expression of homologous recombination (HR)-associated repair genes. This combination presents a potential therapeutic strategy for UVB-induced photodamage and emphasizes the synergistic effects of ROL and RPalm in alleviating UVB-induced DNA damage.

Skin biopsy specimens analyzed included normal skin from five healthy nonatopic adults and both minimally lesional skin and nearby or contralateral nonlesional skin from six adult AD patients.
