The role of microglial lnc-U90926 in ischemic stroke

Project description:Dysregulated long non-coding RNAs (lncRNAs) have been shown to contribute to the pathogenesis of ischemic stroke. However, the potential role of lncRNAs in post-stroke microglial reactivation remains largely unknown. Here, we uncovered that lncRNA-U90926 was significantly increased in the microglia exposed to ischemia/reperfusion in vivo and in vitro. In addition, adenovirus associated virus (AAV)-mediated microglial U90926 silencing alleviated neurological deficits and reduced infarct volume in experimental stroke mice. Microglial U90926 knockdown could reduce the infiltration of neutrophils into ischemic lesion site, which might be attributed to the downregulation of C-X-C motif ligand 2 (CXCL2). Mechanistically, U90926 directly bound to malate dehydrogenase (MDH2) and competitively inhibited MDH2-mediated decay of CXCL2 mRNA. Taken together, our study demonstrated that microglial U90926 aggravated ischemic brain injury via facilitating neutrophil infiltration, suggesting that U90926 might be a potential biomarker and therapeutic target for ischemic stroke.

Repair-Associated Macrophages Increase after Early-phase Microglia Attenuation to Promote Ischemic Stroke Recovery

Project description:<p>Ischemic stroke recovery involves dynamic interactions between the central nervous system and infiltrating immune cells. Peripheral immune cells compete with resident microglia for spatial niches in the brain, but how modulating this balance affects recovery remains unclear. Here, we use PLX5622 to create spatial niches for peripheral immune cells, altering the competition between infiltrating immune cells and resident microglia in male mice following transient middle cerebral artery occlusion (tMCAO). We find that early-phase microglia attenuation promotes long-term functional recovery. This intervention amplifies a subset of monocyte-derived macrophages (RAMf) with reparative properties, characterized by high expression of GPNMB and CD63, enhanced lipid metabolism and pro-angiogenic activity. Transplantation of RAMf into stroke-affected mice improves white matter integrity and vascular repair. We identify Mafb as the transcription factor regulating the reparative phenotype of RAMf. These findings highlight strategies to optimize immune cell dynamics for post-stroke rehabilitation.</p>

Targeting microglial PPARα ameliorates the outcome of ischaemic stroke by enhancing interaction with infiltrating peripheral monocytes/macrophages

Project description:Microglial PPARα downregulation enhances inflammation and worsens ischemic brain injury by promoting Mo/MΦ infiltration, shifting them to a proinflammatory state, and disrupting the BBB. Restoring PPARα expression via AAV transfection reverses these effects, exerting neuroprotection by enhancing microglia–macrophage crosstalk through IL-4 signaling. Targeting microglial PPARα may offer a therapeutic strategy for mitigating acute ischemic stroke.

Inhibition of microglial SLC2A5 attenuates brain injury following acute ischemic stroke [scRNA-Seq]

Project description:The Slc2a5 in microglia plays a crucial role in the brain's injury response. In this study, we utilized single-cell RNA sequencing to examine the impact of Slc2a5 knockout on cell type-specific transcription in the brain. Additionally, we employed single-nucleus RNA sequencing to investigate the cell type-specific transcriptional changes in the brain 24 hours after transient middle cerebral artery occlusion (tMCAO) in response to ischemic stroke. Furthermore, RNA sequencing was conducted to analyze the effects of oxygen-glucose deprivation (OGD) conditions on the transcriptional profiles of wild-type and Slc2a5 knockout microglia in vitro.

Stimulating microglial glucose metabolism and phagocytosis for post-stroke remyelination and cognitive function recovery

Project description:New research shows that disease-associated microglia (DAM) in neurodegenerative brains present features of elevated phagocytosis, lysosomal functions, and lipid metabolism, which benefit brain repair. The underlying mechanisms remain poorly understood. Intracellular pHi is important for regulating aerobic glycolysis in microglia, where Na/H exchanger (NHE1) is a key pH regulator by extruding H+ in exchange of Na+ influx. We report here that post-stroke Cx3cr1-CreER+/-;Nhe1flox/flox (Nhe1 cKO) brains displayed stimulation of microglial transcriptomes of rate-limiting enzyme genes for glycolysis, tricarboxylic acid cycle, and oxidative phosphorylation (OXPHOS). The other upregulated genes included the DAM hallmark genes (Apoe, Trem2, Spp1) as well as genes for phagocytosis and LXR/RXR pathway activation. The cKO microglia exhibited increased OXPHOS capacity and higher phagocytic activity, which led to enhanced synaptic pruning, oligodendrogenesis, and remyelination. This study reveals that genetic blockade of microglial NHE1 stimulated glucose immunometabolism to support phagocytosis function for tissue remodeling and post-stroke cognitive function recovery.
