Prefrontal cortex bulk RNA-seq analysis of drug-treated PTEN KI heterozygous mice at P40

ABSTRACT: Autism spectrum disorder (ASD) affects approximately 2.8% of the population and is characterized by substantial genetic heterogeneity. Bulk RNA sequencing was performed on prefrontal cortex tissue from PTEN KI heterozygous (HT) and wild-type (WT) mice. Animals were analyzed at postnatal day 40 (P40) following three weeks of treatment with fluoxetine or lithium. Both male and female mice were included. Dissected prefrontal cortex tissue was used for RNA extraction and sequencing.

Project description:Autism spectrum disorders (ASD) affect ~2.8% of the population, but the underlying biological mechanisms remain poorly understood, largely due to significant genetic heterogeneity. Mouse models have identified numerous candidate mechanisms; however, whether these can be grouped into functionally coherent subtypes remains unclear, limiting targeted, mechanism-based treatments. We analyzed prefrontal cortex transcriptomes from 17 distinct ASD mouse models, encompassing both sexes, under baseline and two separate drug-treatment conditions, totaling 1,008 mice across 204 groups.
