Tet2 deficiency alters CD4+ T-cell function and promotes T-cell lymphoma with a TFH-cell immunophenotype.

ABSTRACT: TET2 mutations are frequent in TFH derived lymphomas, but how epigenetic disruption initiates malignant T cell transformation is unclear. We performed CRISPER mediated TET2-knockout in normal human CD4+ T cells and culture them long term. We perfromed scRNAseq after 40 or 90 days or long-term culture (>200 days). ScRNA-seq revealed a persistent proliferative cluster characterized by elevated stem like transcriptional features compared to wild type counterparts
