Effect of ARNT on hypoxic response in Panc1 cells

ABSTRACT: The hypoxic response is influence by both the tissue of origin and local environmental influences. The hypoxic response is mediated in large part by the hypoxia-inducible transcription factors HIF-1 and HIF-2, which share a common beta subunit (also called ARNT). We looked at Panc1 cells, a pancreatic adenocarcinoma cell line, and compared the response to hypoxia in both regular tissue culture media and in pancreatic TIFM (tumor interstitial fluid media), and assessed the degree to which ARNT knockout mediates the observed changes

Project description:In this study, we provide new insight into the role of the hypoxia-inducible factor (HIF) complex in the inhibition of syncytiotrophoblast formation under low oxygen. Through a genome-wide analysis of binding sites for the aryl hydrocarbon nuclear receptor translocator (ARNT; a key component of HIF), we identified the downstream targets of ARNT/HIF that potentially mediate its repressive effect on syncytialization.

Project description:We used hydrogen-deuterium exchange mass spectrometry (HDX-MS) to investigate the influence of OEA on HIF-3A-ARNT.HDX-MS data indicated that OEA binding stabilized multiple residues within the Gβ and Fα regions of HIF-3A PAS-B domain.It is possible that the stabilized Gβ and Fα function as an “interlayer” to further contact with the AB-loop of ARNT.

Loss of ARNT Impair Adaptability of Muscular Exercise in Aged Mice

Project description:The adaptation of regimented exercise in skeletal muscle including muscular hypertrophy and enhanced strength decline significantly with aging. Transcriptome analysis following RNA sequencing reveals extensive activation of hypoxia-related genes in young exercised mice versus the sedentary, but absent in aged exercised mice. Particularly, less expression of aryl hydrocarbon receptor translocator (ARNT) was observed in response to exercise in aged mice. Young mice underwent skeletal muscle-specific knockout of ARNT (ARNT mKO) obtain deficient benefit from exercise resembling the aged mice. The deletion of ARNT associated with decreased expression of Notch1 intracellular domain(N1ICD) impair muscle hypertrophy and regeneration. Administration of ML228, a systematic agonist of ARNT, rescued skeletal muscle adaptabilities in old mice, which was suppressed by administrating N1ICD inhibitor(DAPT). These results suggest that the loss of skeletal muscle ARNT is partially responsible for diminished response to exercise in aging and activation of hypoxia signaling holds promise for rescuing the adaptability in aged muscle.

Characterisation of HIF-dependent alternative isoforms in pancreatic cancer

Project description:Intra-tumor hypoxia is a common feature in many solid cancers. Although transcriptional targets of hypoxia-inducible factors (HIFs) have been well characterized, alternative splicing or processing of pre-mRNA transcripts which occurs during hypoxia and subsequent HIF stabilization is much less understood. Here, we identify HIF-dependent alternative splicing events after whole transcriptome sequencing in pancreatic cancer cells exposed to hypoxia with and without downregulation of the aryl hydrocarbon receptor nuclear translocator (ARNT), a protein required for HIFs to form a transcriptionally active dimer.

Transcription profiling of human drug-resistant Panc1/mock cells and Panc1 cells expressing wildtype syk

Project description:We recently identified the nonreceptor tyrosine kinase syk as a tumor suppressor in pancreatic ductal adenocarcinoma cells. Reintroduction of syk into Panc1 cells promoted a more differentiated phenotype and retarded invasion and tumorigenic growth. Gene array analysis identified over 2,000 transcripts differentially expressed at FDR<0.01. Among these were members of the MMP2 axis, which were subsequently shown to regulate Panc1 invasion. Experiment Overall Design: Affymetrix global gene arrays were used to analyse differences in gene expression patterns in Panc1 cells stably reexpressing syk, or vector-only mock controls. RNA was harvested from cells grown under identical conditions in standard culture.
