Project description:It is generally accepted that hepatic gluconeogenesis, the synthesis of glucose from other carbon substrates (i.e.: glycerol, pyruvate, amino acids), is active in the fasted state and inactive in the fed state. In contrast, de novo lipogenesis (synthesis of fatty acids) is active in the fed state and is inactive in the fasted state. However, RNAPII ChIP-seq demonstrated that periportal but not pericentral hepatocytes display a basal (fed state) transcriptional activation of gluconeogenic genes that correlated with the basal level of gluconeogenic mRNA expression. Targeted single cell RNA-seq, HCR RNA-FISH, and PrimeFlow identified a subpopulation of periportal hepatocytes (approximately 10% of total hepatocytes) that simultaneously co-express both gluconeogenic and lipogenic genes in the fed state. These dual-positive hepatocytes identified in normal mouse livers are also present in human hepatocytes from humanized mouse livers. Spatial metabolic imaging coupled with stable isotope tracing analyses at the single cell level demonstrated that these individual hepatocytes simultaneously undergo both gluconeogenesis and de novo lipogenesis. Euglycemic-hyperinsulinemic clamps coupled with PrimeFlow analyses further demonstrated that this novel hepatocyte subpopulation is naturally insulin resistant. Moreover, high-fat diet-induced insulin resistance and hepatic steatosis were accompanied by an increase in the number of these dual-positive hepatocytes, suggesting a paradigm shift in our understanding of how the liver becomes insulin resistant. Taken together, these data demonstrate the existence of a unique, naturally insulin-resistant subset of mouse and human periportal hepatocytes that have the capacity to simultaneously undergo both gluconeogenesis and de novo lipogenesis.

Project description:The liver acts as a master regulator of metabolic homeostasis in part by performing gluconeogenesis. This process is dysregulated in type 2 diabetes, leading to elevated hepatic glucose output. The parenchymal cells of the liver (hepatocytes) are heterogeneous, existing on an axis between the portal triad and the central vein, and perform distinct functions depending on location in the lobule. Here, using single cell analysis of hepatocytes across the liver lobule, we demonstrate that gluconeogenic gene expression (Pck1 and G6pc) is relatively low in the fed state and gradually increases first in the periportal hepatocytes during the initial fasting period. As the time of fasting progresses, pericentral hepatocyte gluconeogenic gene expression increases, and following entry into the starvation state, the pericentral hepatocytes show similar gluconeogenic gene expression to the periportal hepatocytes. Similarly, pyruvate-dependent gluconeogenic activity is approximately 10-fold higher in the periportal hepatocytes during the initial fasting state but only 1.5-fold higher in the starvation state. In parallel, starvation suppresses canonical beta-catenin signaling and modulates expression of pericentral and periportal glutamine synthetase and glutaminase, resulting in an enhanced pericentral glutamine-dependent gluconeogenesis. These findings demonstrate that hepatocyte gluconeogenic gene expression and gluconeogenic activity are highly spatially and temporally plastic across the liver lobule, underscoring the critical importance of using well-defined feeding and fasting conditions to define the basis of hepatic insulin resistance and glucose production.

Project description:The role and molecular mechanisms of intermittent fasting (IF) in non-alcoholic steatohepatitis (NASH) and its transition to hepatocellular carcinoma (HCC) are unknown. Here, we identified that an IF 5:2 regimen (two non-consecutive days of food deprivation per week), initiated in the active phase of mice, prevents NASH development as well as ameliorates established NASH and fibrosis without affecting total calorie intake. Furthermore, the IF 5:2 regimen also blunted NASH-HCC transition when applied therapeutically in two independent models of diet-induced NASH and NASH-HCC. The timing (7pm-7pm > 7am-7am), length (24h > 12h) and number (5:2 > 6:1) of fasting cycles as well as the type (WD / CDHFD) of NASH diet were all critical parameters determining the effectiveness of the fasting benefits. Combined proteome, transcriptome and metabolome analyses identified that PPARα and glucocorticoid signalling-induced phosphoenolpyruvate carboxykinase 1 (PCK1) act co-operatively as hepatic executors of the fasting response by promoting fatty acid catabolism and gluconeogenesis while suppressing anabolic lipogenesis. In line, PPARα targets and PCK1 were reduced in human NASH. Notably, only fasting during the active phase (7pm-7pm) of mice robustly induced glucocorticoid signalling in hepatocytes including PCK1 expression; while both active and inactive phase (7am-7am) fasting induced free fatty acid-induced PPARα signalling, highlighting the increased efficacy of fasting during the active phase. However, hepatocyte-specific glucocorticoid receptor (GR) deletion only partially abrogated the hepatic fasting response, illustrating the need for both glucocorticoid-induced PCK1 expression and free fatty acid-induced PPARα activation for mediating the benefits of fasting. In support, the combined knockdown of Ppara and Pck1 in vivo abolished the beneficial outcomes of fasting against inflammation and fibrosis, confirming their causal relationship in integrating systemic signalling in hepatocytes. Moreover, overexpression of Pck1 alone or together with Ppara in vivo lowered hepatic triglycerides and steatosis upon Western-diet feeding. Altogether, our data support that the IF 5:2 regimen could be a promising and viable intervention against NASH and subsequent liver cancer.

Project description:SEPHS2, a key enzyme in synthesizing selenoproteins, which converts hydrogen selenide into selenophosphate. We found that SEPHS2 knockout inhibits oxidative phosphorylation (OXPHOS), but has no effect on glycolysis, as indicated by an seahorse assay. SEPHS2 loss redirects more glucose to the pentose phosphate pathway (PPP). Mechanistically, SEPHS2 knockout elevates NAD+ levels, which are essential for Sirtuin activity. The increase in the activity of SIRT2 leads to deacetylation of PCK1, a posttranslational modification required to prevent PCK1’s ubiquitination and subsequent degradation. Thus, SEPHS2 knockout stabilizes PCK1 by reducing its ubiquitination. Since PCK1 is a key regulator of gluconeogenesis, SEPHS2 knockout promotes gluconeogenesis while reducing cellularglucose uptake, down-regulating oxidative phosphorylation and enhancing the PPP.

Project description:Metabolic dysregulation may serve as a pivotal driver of inflammatory bowel disease (IBD), with the crosstalk between metabolism and the immune system emerging as a critical research focus. Ulcerative colitis (UC) patients exhibit enhanced glycolysis but suppressed gluconeogenesis in the intestinal mucosa, yet the mechanistic links between these metabolic alterations and mucosal injury remain poorly defined. This study establishes phosphoenolpyruvate carboxykinase 1 (Pck1), a key rate-limiting enzyme in gluconeogenesis, as a central regulator of UC progression. We found Pck1 expression was significantly downregulated in colon tissues of UC patients and mice with DSS‑induced colitis. Utilizing an intestinal epithelial cell (IEC)-specific Pck1 knockout mouse model, we demonstrated for the first time that Pck1 deficiency exacerbates intestinal inflammation. Mechanistically, Pck1 depletion induces metabolic reprogramming that upregulates Pla2g6 expression, driving abnormal hydrolysis of phosphatidylcholine and phosphatidylethanolamine in IECs. Pharmacological inhibition of Pla2g6 ameliorated colitis in Pck1-deficient mice. These findings position Pck1 as a potential early diagnostic biomarker for UC and suggest that therapeutic targeting of the Pck1-Pla2g6 metabolic axis may yield novel strategies for UC management.
