ABSTRACT: Photorhabdus luminescens bacteria alternate lifestyles between its pathogenic insect host and its mutualistic nematode host. We found that Photorhabdus changes radically from the insect pathogenic form (P-form) to slow growing small cells when initiating mutualism (M-form). Here we characterize the gene expression of the M-form relative to the P-form.

Project description:Photorhabdus luminescens bacteria alternate lifestyles between its pathogenic insect host and its mutualistic nematode host. We found that Photorhabdus changes radically from the insect pathogenic form (P-form) to slow growing small cells when initiating mutualism (M-form). Here we characterize the gene expression of the M-form relative to the P-form. RNA was extracted from mid-exponential phase bacterial cultures of genetically locked P-form and M-form cells in LB media supplemented with 1 g/liter sodium pyruvate at 28 degree celcius. M-form gene expression was calculated relative to P-form. Three replicates were performed for each biological sample.

RNA-Sequencing analysis of the Drosophila transcriptomic response to infection by entomopathogenic nematodes and their mutualistic bacteria

Project description:We report the application of next generation RNA sequencing to analyze the transcriptional response of Drosophila adult flies to infection by the insect pathogenic nematodes Heterorhabditis bacteriophora and their mutualistic bacteria Photorhabdus luminescens, either separately or together. We find that Heterorhabditis and Photorhabdus differentially modulate a large number of genes, many of which participate in metabolic functions, stress responses, repression of gene transcription and neuronal activities. We have also identified Drosophila genes with potential role in nematode recognition and others with putative anti-nematode properties. These findings generate novel insights into how the host immune function is shaped to respond against nematode parasites and their associated bacteria.

Project description:Photorhabdus luminescens lives in a mutualistic association with entomopathogenic nematodes and is pathogenic for insects. Variants of Photorhabdus frequently arise. VAR* is a colonial and phenotypic variant displaying delayed pathogenicity in the insect, Spodoptera littoralis. In this study, we evaluated the role of transcriptomic modulation in determining the phenotypic variation and delayed pathogenicity of VAR* with respect to the corresponding wild-type form, TT01alpha. A P. luminescens microarray was used to determine changes in transcript profiles in the exponential and stationary phase growth phase of VAR* and TT01alpha grown in LB broth. Direct transcriptomic comparisons were carried out between VAR* and TT01alpha grown in LB broth. RNA were extracted from bacterial cells in the exponential growth phase (6-hour-old culture, 5 x 107 cfu/ml) and stationary phase (45-hour-old culture, 5 x 108 cfu/ml). Each microarray comparison (TT01alpha versus VAR* growth in exponential growth or stationary phase) included six slides, with three dye-swapping replicates. Each microarray comparison is done twice with two independant biological extractions (repbiol1 or 2).

DNA adenine methyltransferase (Dam) overexpression impairs Photorhabdus luminescens motility and virulence

Project description:Dam, the most described bacterial DNA-methyltransferase, is widespread in gamma-proteobacteria. Dam DNA methylation can play a role in various genes expression and is involved in pathogenicity of several bacterial species. In the entomopathogenic bacterium Photorhabdus luminescens, a dam ortholog was identified. Overexpression of dam in P. luminescens did not impair growth ability in vitro. In contrast, compared to a control strain harboring an empty plasmid, a significant decrease in motility was observed in the dam-overexpressing strain. In addition, the dam-overexpressing P. luminescens strain showed a delayed virulence compared to that of the control strain after injection in larvae of the lepidopteran Spodoptera littoralis. These results reveal that Dam plays a major role during P. luminescens insect infection.

Controlling a Necessary Evil: Quorum Sensing Attenuates Virulence in Sodalis praecaptivus.

Project description:Sodalis praecaptivus is a close relative and putative environmental progenitor of the widely distributed, insect-associated, Sodalis-allied symbionts. Mutant strains of S. praecaptivus, lacking critical components of a quorum sensing (QS) apparatus acquire a rapid and potent killing phenotype following microinjection into an insect host. Transcriptomic and genetic analyses indicate that insect killing occurs as a consequence of virulence factors, including insecticidal toxins and enzymes that degrade the insect integument that are normally repressed by QS at high infection densities. This unorthodox method of regulation ensures that virulence factors are only utilized to initiate the insect-bacterial association. Once bacteria reach a sufficient infection density in host tissues, the QS circuit represses the expression of these harmful genes, facilitating a long-lasting and benign association. We discuss the implications of the functionality of this QS system in the context of the establishment and evolution of mutualistic relationships involving these bacteria.

Molecular causes of an evolutionary shift along the parasitism-mutualism continuum in a bacterial symbiont

Project description:Symbiosis is a ubiquitous phenomenon in nature, and these inter-species interactions have a massive impact on organisms, shaping the world around us today. The relationship between the partners in microbial symbioses have been described as existing along a parasitism-mutualism continuum, and the dynamics of this continuum are dependent upon numerous genotypic and environmental factors. Theoretical and experimental studies show that vertical transmission (VT) leads to the evolution of mutualistic traits, whereas horizontal transmission (HT) facilitates the emergence of parasitic features. However, these studies focused on phenotypic data, and we know little about underlying molecular changes at the genomic level. Here we show that a dramatic shift in the frequency of genetic variants, coupled with major changes in gene expression, allow an obligate intracellular bacterial symbiont to alter its position in the mutualism-parasitism continuum depending on the mode of between-host transmission. We found that increased virulence in horizontally transmitted chlamydiae residing in amoebae was a result of processes occurring at the infectious stage of the chlamydia’s developmental cycle. Specifically, genes involved in energy production required for extracellular survival as well as the type III secretion system (T3SS) - the symbiont’s primary virulence mechanism - were significantly upregulated. Our results identify the genomic and transcriptomic dynamics sufficient to favor parasitic or mutualistic strategies.

Fig and fig wasp mutualism

Project description:A striking property of the ancient and obligate mutualism between figs and their pollinating wasps is that fig wasps consistently oviposit in the inner flowers of the fig syconium (gall flowers, which develop into galls that house developing larvae), but typically do not use the outer ring of flowers (seed flowers, which develop into seeds). To better understand differences between gall and seed flowers that might influence oviposition choices, and the unknown mechanisms underlying gall formation, we used a metatranscriptomic approach to analyze eukaryotic gene expression within fig flowers at the time of oviposition choice and early gall development. Consistent with the unbeatable seed hypothesis, which posits that only a portion of fig flowers are physiologically capable of responding to gall induction or supporting larval development, we found significant differences in gene expression assigned to defense and metabolism between gall- and seed flowers in receptive syconia. Transcripts assigned to flavonoids and defense were especially prevalent in receptive gall flowers, and carbohydrate metabolism was significantly up-regulated relative to seed flowers. In turn, high expression of the venom gene icarapin during wasp embryogenesis within galled flowers distinguishes it as a candidate gene for gall initiation. In response to galling, the fig significantly up-regulates the expression of chalcone synthase, which previously has been connected to gall formation in other plants. This study simultaneously evaluates the gene expression profile of both mutualistic partners in a plant-insect mutualism and provides evidence for a stability mechanism in the ancient fig-fig wasp association. We examined two different Ficus flower types at two different time points. Each sample contained a pool of hundreds of individual flowers from multiple sycomia.
