The sampling time window covered from the earliest day that stomach sampling is anatomically feasible to the latest weeks when stomachs are close to mature. We applied a fast-seq proteomics workflow, a label-free quantitative proteomics approach in combination with small scale reverse phase pre-fractionation strategy.

Gene expression profile of the stomach from an AGR2 null mouse

Project description:The in vivo function of AGR2 was examined by generating an AGR2 null mouse. The mice died prematurely due to hyperplasia and dysplasia of the glandular stomach. Gene expression profiling was performed to compare differences in transcription between wild-type and null AGR2 mice. Female AGR2 wild-type and knockout mice were sacrificed at the age of 26 weeks and the glandular stomachs were removed. Total RNA was isolated using TRIzol reagent (Invitrogen, Carlsbad, CA). The RNA was was reverse transcribed, labeled, and hybridized to mouse oligonucleotide DNA microarrays (Illumina MouseRef-8 v2 Expression BeadChip)

Expression data from stomach of germ-free and gnotobiotic mice

Project description:The aim of this study was to examine the role of indigenous lactobacilli in the physiological development of the stomach in mice using microarray analysis. In lactobacilli-associated gnotobiotic mice, an increased expression of the genes related to the muscle system development, such as nebulin and troponin, was observed. On the other hand, the expression of the gastrin gene dramatically decreased. A microarray analysis of the stomachs infected with H. pylori also showed both the up-regulation of muscle cell genes and the down-regulation of gastrin genes. Male germfree BALB/c mice were orally inoculated with 109 CFU lactobacilli and their stomachs were excised after 10 days for analyses. In the other experiment, 8-week-old mice were orally inoculated using a stomach tube on three consecutive days with 109 CFU of H. pylori. The stomachs of the germfree mice were used as controls. For the DNA microarray analysis, RNA was prepared from the stomach and then applied to a genome array.

Lgr5-Expressing Chief Cells Drive Epithelial Regeneration and Cancer in the Oxyntic Stomach

Project description:Adult stem cells residing within tubular glands of the corpus epithelium are believed to fuel daily tissue renewal, but their identity remains controversial. Lgr5, marks both homeostatic stem cells and “reserve” stem cells in multiple tissues. Here, we report Lgr5 expression in a subpopulation of chief cells in mouse and human corpus glands. Using a new, non-variegated Lgr5-2A-CreERT2 mouse model, we show by cell fate mapping that Lgr5-expressing chief cells do not behave as corpus stem cells under homeostatic conditions, but are recruited to function as stem cells to effect epithelial renewal following injury. In vivo ablation of the Lgr5+ cells severely impairs epithelial homeostasis in the corpus, indicating an essential role in maintaining the resident stem cell pool. We additionally define the Lgr5+ chief cells as a major cell of origin of gastric metaplasia. These findings reveal clinically relevant insights into tissue homeostasis, repair and cancer in the corpus.

Transcriptional profiles of stomach organoids from human embryonic stem cells.

Project description:Gastric ulcer, which affect many of patients and is deeply related with gastric cancer, is caused by chronic gastric acid stimulation. Stomach fundus, the main body of stomach, is a major source of gastric acid and peptidase for food digestion. Recapturing the main body of stomach requires mainly 3 functionally differentiated cells; parietal (oxyntic) cells, chief (zymogenic) cells, and surface mucous foveolar (pit) cells. We have previously shown the induction of stomach tissue with functional secreting activities by directed differentiation of mouse embryonic stem cells (ES cells) to stomach primordium with both gut epithelium and splanchnic mesoderm. However, generating human stomach with fundus and such functions has been elucidated and a long-desired goal. Here, we describe the method for establishing human embryonic stem cell-derived stomach organoids with fundus gland structure. Along with mouse stomach development and de novo stomach generation from mouse ES cells in vitro, we observed gut-like structure formation from human embryonic stem cells by induction of both endoderm and mesoderm. These human embryonic gut could differentiate into stomach primordium by growth factor stimulation as well as stomach development, and form stomach tissue in three-dimensional organoid culture. Furthermore, these stomach organoids contain fundus-like gland with parietal cells and chief cells, some of secreting activities, and is transcriptionally close to human stomach. Human functional stomach derived from embryonic stem cells represent powerful tools for analying human stomach development, and gastric ulcer related disease including gastric tumorgenesis.

Gastric single-cell RNA-sequencing of mice with Helicobacter pylori infection and/or KRAS-driven metaplasia

Project description:Stomachs from mice +/- H. pylori infection, +/- induction of active KRAS in gastric Mist1-expressing cells, were used for gastric single-cell RNA-sequencing. Samples were obtained six and 12 weeks after infection/KRAS induction. Six samples were frozen, then thawed before sequencing (and in some cases pooled from multiple mice) and two samples were freshly prepared and never frozen before sequencing.
