Project description:Using a mouse model of non-small cell lung cancer driven by activated Kras and loss of p53 (KrasG12D;p53fl/fl), we sought to determine which genes change expression in the bulk tumor after repetitive treatment with cispatin. After tumors were treated multiple times with cisplatin and show no further response, we isolated RNA from tumors treated with cisplatin or a vehicle control (PBS) to determine which genes significantly change in expression in chemoresistant bulk tumors.

Project description:In this dataset, we utilized the db/db, uninephrectomy and renin-hypertension mouse model. Bulk RNA-seq on NOD mice prostate tissues treated with etanercept

Project description:Immunohistochemistry studies indicated that etanercept treatment reduced inflammation, epithlial proliferation, and epithelial NFkB activity in NOD prostate tissues compared to control treated mice. A bulk RNA-seq study was conducted on whole mouse prostate tissues from NOD mice treated with PBS control or etanercept for 5 weeks to identify putative pathways involved in these processes. These analyses determined differentially expressed genes and numerous enriched pathways yielding a greater understanding of pathways involved in prostate regression and inhibition of inflammation.

FOS Immunohistochemical Staining of Colorectal Cancer and Its Adjacent Tissues

Project description:Colorectal cancer tissue sections were obtained according to the inclusion criteria. The formalin was used to immersed all cancer specimens. And tissues were cut to 5 μm thickness and placed on glass slides before staining. Endogenous peroxidase activity was inhibited and blocked by de-paraffinizing, rehydrating, and using 5% bovine serum albumin at 37ºC for 30 min. The treated sections were incubated with anti-FOS (promab 30360) at 4ºC overnight and washed three times with PBS. After that, it is required that incubation with secondary anti-peroxidation sunflower at 37ºC for 30 minutes. After washing three times again with PBS, the sections were developed in diaminobenzidine and microscopic images were made by light microscopy.

Bulk RNA-Seq of whole testis sampled from juvenile mice

Project description:Spermatogenesis is a recurring differentiation process that results in the production of male gametes within the testes. During this process, spermatogonial stem cells differentiate to form spermatocytes, which undergo two rounds of meiotic division to form haploid spermatids. Throughout spermiogenesis, round spermatids elongate to form mature sperm. To profile maturing cell types, we generated bulk RNA-seq data from whole testis undergoing the first round of spermatogenesis between post-natal day P6 and P35. We also compared these libraries to adult samples.

Understanding the underlying molecular mechanisms of meiotic arrest in organotypic culture of prepubertal rat testicular tissues

Project description:We report the use of the Bulk RNA-sequencing technology on in vivo and in vitro cultured prepubertal testicular tissues. The goal of this study is to identify the origin of the maturation arrest observed in rat prepubertal testicular tissue cultured with the organ culture system with a transcriptomic approach. We compared the gene expression of 14.5 dpp rat testicular tissues cultured up to 6, 16, 22 and 28 days. The statistical filtration revealed 600 differentially expressed genes showing significant differential abundances in all three comparisons. This study uncovered for the first time several mechanisms that could explain the meiotic arrest observed in cultures of rat prepubertal testicular tissues.

Transcriptomics Analysis of bulk RNA from liver tissues of male mice treated with high-fat diet and formoterol

Project description:To establish the consequences of formoterol treatment on high fat diet-induced liver steatosis and inflammation, we isolated RNA from bulk liver tissue of male C57BL/6J mice that were treated with a high-fat diet for 20 weeks, with treatment with vehicle (DMSO) or formoterol (1 mg/kg) daily for the last 4 weeks of the diet. Mice were euthanized using CO2 and cervical dislocation prior to liver dissection.
