Single-cell RNA-sequencing of Immortalized Alveolar type 2 Cell Lines from Healthy Lungs

ABSTRACT: Using the 10x Genomics platform, we conducted single-cell RNA sequencing to profile our newly immortalized alveolar type 2 (AT2) cell lines. Since primary human AT2 cells are notoriously difficult to propagate long-term, passage repeatedly, and modify genetically, we developed several stable, immortalized lines derived from healthy AT2 cells.

Project description:Background: Mature adipocytes are notoriously difficult to study ex vivo and alternative cell culture systems have therefore been developed. One of the most common models are human adipose progenitor cells (hAPCs). Unfortunately, these display replicative senescence after prolonged culture conditions, which limits their use in mechanistic studies. Methods: Herein, we knocked in human telomerase reverse transcriptase (TERT) into the AAVS1 locus of CD55+ hAPCs derived from abdominal subcutaneous adipose tissue and characterized the cells before and after differentiation into adipocytes. Results: Immortalized TERT-hAPCs retained proliferative and adipogenic capacities comparable to those of early-passage wild type hAPCs for >80 passages. In line with this, our integrative transcriptomic and proteomic analyses revealed that TERT-hAPCs displayed robust adipocyte expression profiles. This was confirmed by functional analyses of lipid turnover where TERT-hAPCs exhibited pronounced responses to insulin and pro-lipolytic stimuli such as isoprenaline, dibutyrul cAMP and tumor necrosis factor alpha. In addition, TERT-hAPCs could be readily cultured in both standard 2D and 3D-cultures and proteomic analyses revealed that the spheroid culture conditions improved adipogenesis. Conclusion: Through descriptive and functional studies, we demonstrate that immortalization of human CD55+ hAPCs is feasible and results in cells with stable proliferative and adipogenic capacities over multiple passages. As these cells are cryopreservable, they provide the additional advantage over primary cells of allowing repeated studies in both 2D and 3D model systems with the same genetic background.

miRNA Expression in Senescent and Immortalized BJ fibroblasts

Project description:Limited data exists regarding changes of microRNA (miRNA) expression during senescence in human cells and no reports correlate telomerase expression with regulation of senescence-related miRNAs. We used miRNA microarrays to provide a detailed account of miRNA profiles for early passage and senescent human foreskin (BJ) fibroblasts as well as early and late passage immortalized fibroblasts (BJ-hTERT) that stably express the human telomerase reverse transcriptase subunit hTERT. The revelation that miRNA expression changes with extended passaging in BJ-hTERT cells will contribute to a comprehensive understanding of the connections between telomerase expression, senescence and processes of cellular aging.

scRNAseq on mouse lungs of bleomycin injured aged mice

Project description:Pulmonary fibrosis remains a significant public health concern due to an increasing incidence and limited treatment options. Alveolar type 2 epithelial cell (AT2) senescence causes stem cell exhaustion and progenitor failure, leading to ineffective and aberrant alveolar repairs and unresolved fibrosis. AT2 senescence regulatory molecules require additional studies. We demonstrate that the overexpression of syndecan-1 on IPF-AT2 cells is positively associated with the AT2 composite transcriptomic senescence score (SenMayo Senescence Score) and cellular senescence proteins. These findings imitate those of young and aged bleomycin-injured wild-type (WT) mice. The roles of syndecan-1 in regulating AT2 cellular senescence are corroborated in human and mouse lung epithelial cell lines. Functionally, the AT2 alveolospheres from bleomycin-injured Sdc1–/– mice had higher colony-forming efficiency (CFE) and normal differentiation, whereas WT alveolospheres were less proliferative and aberrant differentiation. Using precision-cut lung slices (PCLS), bleomycin-injured WT lungs had lower AT2 cells and a lower secretory surfactant protein C than Sdc1–/– PCLS, indicating the detrimental role of excess syndecan-1. We further showed that the transcriptomic p53 activity score is higher in IPF and bleomycin-injured WT AT2 cells compared to Sdc1–/– AT2 cells. Additionally, syndecan-1 activated p53 by inducing acetylation at lysine379. Our work indicates that syndecan-1 reprograms AT2 cells to become senescent during lung fibrogenesis and likely through the p53 signaling pathway.

Project description:Background: MLL (KMT2A)-EB1 (MAPRE1) fusion was identified in a patient with de novo pro-B acute lymphoblastic leukemia. To investigate the leukemogenesis of MLL-EB1 fusion, a retroviral transduction of MLL-EB1 to murine bone marrow cells was performed. A frequent MLL fusion, MLL-AF10(OM-LZ), was used as a positive control. Results: Two MLL-EB1 immortalized cell lines (ME1 and ME2G), and a MLL-AF10(OM-LZ) immortalized cell line (12G) were generated. Microarray results showed that many genes including Evi1 and Ets1 were differentially expressed in ME1/ME2G and 12G cell lines.
