A Comparative Analysis of the Methylation Status of Non-Coding RNA promoters in Fibroid and Matched Myometrium

ABSTRACT: Genomic DNA from 8 paired uterine leiomyoma (fibroid) and matched myometrial tissues was analyzed using the Arraystar Human ncRNA Promoter Array (4×180K) combined with methylated DNA immunoprecipitation (MeDIP-chip). This platform interrogates promoter regions of long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) to identify DNA methylation enrichment patterns. Differential promoter methylation analysis identified 538 lncRNAs and 61 miRNAs with significant differences between fibroid and matched myometrium. Among these, 300 lncRNAs and 47 miRNAs showed increased promoter methylation, whereas 238 lncRNAs and 14 miRNAs showed decreased promoter methylation in fibroid tissues.
