M6Am methyltransferase PCIF1 regulates osteogenesis via WNT signaling

ABSTRACT: Mesenchymal stem cells (MSCs) were isolated from Prrx1-Cre;Pcif1fl/fl mice and Pcif1fl/fl littermates and cultured in osteogenic medium for 7 days to investigate the role of Pcif1 in osteogenic differentiation. Total RNA was extracted using Trizol reagent and purified with poly-T oligo-attached magnetic beads. Sequencing libraries were prepared and sequenced on an Illumina PE150 platform. Raw reads were processed and mapped to the Mus musculus reference genome (mm10) using HISAT2. Differential gene expression analysis was performed with the DESeq2 R package using thresholds of |fold change| ≥ 2 and adjusted p-value ≤ 0.05. Gene Ontology (GO) and KEGG pathway enrichment analyses were conducted using the clusterProfiler R package to identify biological processes and pathways affected by Pcif1 deletion during osteogenic differentiation. This dataset provides insights into the transcriptional programs regulated by Pcif1 in MSC osteogenesis.
