Single-cell RNA sequencing of chondrocytes from osteoarthritis (OA) patients and healthy controls

ABSTRACT: Osteoarthritis (OA) is a common degenerative joint disease characterized by progressive cartilage degeneration and osteophyte formation. However, the mechanisms underlying OA progression remain poorly understood. Single-cell RNA sequencing (scRNA-seq) was performed on chondrocytes from three OA patients undergoing total knee arthroplasty and three control donors undergoing arthroscopic surgery for anterior cruciate ligament (ACL) injury to explore cellular heterogeneity and dysregulated pathways. These data provide a resource for investigating the molecular mechanisms underlying OA progression.

Single-cell RNA-Seq reveals changes in immune landscape in post-traumatic osteoarthritis

Project description:Osteoarthritis (OA) is the most common joint disease, affecting over 300 million people world-wide. Accumulating evidence attests to the important roles of immune system in OA pathogenesis. Understanding the role of various immune cells in joint degeneration or joint repair after injury is helpful for improving therapeutic strategies for treating OA. Post-traumatic osteoarthritis (PTOA) develops in ~50% of individuals who have experienced an articular trauma like an anterior cruciate ligament (ACL) rupture. Here, using highly sensitive single-cell RNA sequencing technology, we delineated the temporal dynamics of immune cell accumulation in the mouse knee joint after ACL rupture.

Comparative Transcriptomics of STR/ort, C57BL/6 and MRL/MpJ Knee Joints

Project description:Injuries to the anterior cruciate ligament (ACL) often result in post-traumatic osteoarthritis (PTOA). PTOA accounts for ~12% of all osteoarthritis (OA) cases, yet the mechanisms contributing to OA after joint injury are not well understood. To better understand the molecular mechanisms behind PTOA development following ACL injury, we profiled ACL injury-induced gene expression changes in knee joints of three mouse strains with varying susceptibility to PTOA: STR/ort (highly susceptible), C57BL/6 (moderately susceptible) and super-healer MRL/MpJ (not susceptible) and identified genes differentially expressed between these strains at 0-day [before injury], 1-day, 1-week, and 2-weeks post-injury. This study highlights many new potential therapeutic targets and OA biomarkers.

Effect of donor age on the proportion of mesenchymal stem cells derived from anterior cruciate ligaments

Project description:Analysis of gene expression profiles of ACL-MSCs derived from total knee arthroplasty (TKA) and ACL reconstruction patients. We hypothesized that the proportion of MSCs in ACL samples correlates negatively with donor age, i.e., the proportion of MSCs in ACL samples are higher in younger patients undergoing ACL reconstruction than in older patients undergoing TKA. Results provide additional information on the the effect of age on MSC properties, in particular, the differential regulation of genes encoding components of the extracellular matrix (ECM).

Single-cell sequencing of murine articular cartilage

Project description:Osteoarthritis (OA) is a degenerative joint disease characterized by progressive cartilage loss, bone remodeling, synovial inflammation, and significant joint pain, often resulting in disability. Injury to the synovial joint such as the anterior cruciate ligament (ACL) tear is the major cause of OA in young adults. Currently, there are no approved therapies available to prevent joint degeneration or rebuild articular cartilage destroyed by OA, primarily because our understanding of the cellular and molecular changes that contribute to joint damage is very limited. The synovial joint is a complex structure composed of several tissues including articular cartilage, subchondral bone, synovium, synovial fluid, and tensile tissues including tendons and ligaments. In the present study, using single-cell RNA sequencing (scRNA-seq), we examined the cellular heterogeneity in articular cartilage from mouse knee joints and determined the knee joint injury-induced early molecular changes in the chondrocytes that could contribute to OA.

Project description:Knee ligaments, the anterior cruciate ligament (ACL) and medial collateral ligament (MCL), have unique functions but distinct regeneration capacities. However, the landscape of cellular heterogeneity in ligaments remains unclear. Here we profile the transcriptional signatures of ACL and MCL cells using single-cell RNA-sequencing. These ligaments consist of three fibroblast types that express Col22a1, Col12a1, or Col14a1 but have distinct localizations in the tissue. In addition, we find substantial heterogeneity in Col12a1 and Col14a1 positive cell clusters when comparing the ACL and MCL. Furthermore, we identified aging related changes in both cell composition and gene expression in ligaments.

RNA-Seq of anterior cruciate ligament and knee joint synovium in dogs that are affected with cruciate rupture compared to matched controls.

Project description:The intent of the experiment was to identify genes that were differentially expressed between dogs affected with anterior cruciate ligament (ACL) rupture and breed-matched controls. Anterior cruciate ligament and knee synovial tissue biopsies were collected from 4 ACL rupture affected cases and 4 unaffected control dogs. Cases and controls were matched as closely as possible based on breed, sex, neutered status, age, and weight. Medications that the dogs were taking at the time of sample collection were also considered. We prioritized sample size and quality above all other variables, therefore, two matched pairs of Golden Retrievers were chosen with two matched pairs of Labrador Retrievers for this analysis. Tissues from cases were collected during knee stabilization surgery. Tissues from unaffected control dogs were collected from dogs undergoing pelvic limb amputation or euthanasia for reasons unrelated to this study. Illumina TruSeq RNA libraries were constructed and 150bp paired-end sequencing was performed using the Illumina Hi-Seq 2500 platform. Table 1.
