Murine Dorsal Root Ganglion Single Cell Sequencing of Sham Intrafemoral Inoculation

ABSTRACT: We have employed a single cell sequencing approach using 10x Genomics scRNAseq to study murine dorsal root ganglion (DRG) sensory neurons contribution to cancer-induced bone pain. DRG are the hub of sensory neurons and pain signaling. These findings establish a non-tumor-bearing reference dataset.

Comprehensive analysis of Long non-coding RNA expression in dorsal root ganglion reveals cell type specificity and dysregulation following nerve injury [human iPS]

Project description:Dorsal root ganglion (DRG) neurons provide connectivity between peripheral tissues and spinal cord. Transcriptional plasticity within DRG sensory neurons after peripheral nerve injury contributes to nerve repair but also leads to maladaptive plasticity, including the development of neuropathic pain. This study presents tissue and neuron specific expression profiling of both known and novel Long Non-Coding RNAs (LncRNAs) in rodent DRG following nerve injury. We have identified a large number of novel LncRNAs expressed within rodent DRG, a minority of which were syntenically conserved between mouse and rat and which including both- intergenic and antisense LncRNAs. We have also identified neuron type-specific LncRNAs in mouse DRG, and LncRNAs that are expressed in human IPS cell-derived sensory neurons. We show significant plasticity in LncRNA expression following nerve injury, which in mouse is strain dependant. This resource is publicly available and will aid future studies of DRG neuron identity and the transcriptional landscape in both naïve and injured DRG.

Project description:The molecular identity of touch sensation is still largely unknown. We choose an extrem and very specific example, touch sensation of pennis glan, to study this problem. It's known from previous retro-grade labeling result that only one DRG in rat innervate the pennis glan, which makes it idea for microarry analysis. To clone the mechanosensory receptor specifically or highly expressed in the primary sensory neurons innervating pennis glan. There is a specific mechanosensory receptor in the primary sensory neurons innervating pennis glan. 1. Retrograde labeling the innervating nerve by DiI 2. Dissect DRGs, dissociate neurons and pick up the fluorescent cells. 3. Single nucleus transcriptomic analysis of human dorsal root ganglion neurons

Project description:Somatosensory neurons with cell bodies in the dorsal root ganglia (DRG) project to the skin, muscles, bones, and viscera to detect touch and temperature as well as to mediate proprioception and many types of interoception. In addition, the somatosensory system conveys the clinically relevant noxious sensations of pain and itch. Here we used single nuclear transcriptomics to characterize the classes of human DRG neurons that detect these diverse types of stimuli. Notably, multiple types of human DRG neurons have transcriptomic features that resemble their mouse counterparts although expression of genes considered important for sensory function often differed between species. More unexpectedly, we demonstrated that several classes of mouse neurons have no direct equivalents in humans and human specific cell-types were also identified. This dataset should serve as a valuable resource for the community, for example as means of focusing translational efforts on molecules with conserved expression across species.

Gene expression profiles from multimodal patch-seq neuron samples of the pig dorsal root ganglion

Project description:This dataset comprises single-cell transcriptomic profiles derived from the dorsal root ganglia (DRG) of pigs, aimed at elucidating the molecular identity of mechano-insensitive C-fibres (CMis) implicated in neuropathic pain. Using an innovative approach that combines electrophysiological characterization with Patch-seq, we have successfully captured and analyzed the transcriptomes of primary sensory neurons. These neurons were characterized based on their electrophysiological properties, mirroring those of human CMis, which play a pivotal role in the persistence of neuropathic pain. The presented dataset enriches our understanding of the cellular and molecular landscape of neuropathic pain but also opens new avenues for the development of targeted therapeutic interventions. This dataset is expected to serve as a valuable resource for researchers in the fields of sensory biology and pain neuroscience.

Project description:Sensory neurons of the dorsal root ganglion (DRG) play a crucial role in maintaining tissue homeostasis by sensing and initiating responses to stimuli. While most preclinical studies of DRGs are conducted in rodents, much less is known about the mechanisms of sensory perception in primates. We generated a transcriptome atlas for mouse, guinea pig, cynomolgus monkey, and human DRGs using a framework that implements a common laboratory workflow (i.e sequencing technologies, laboratory protocols, and sample archival methods) and multiple data-integration approaches to generate accurate cross-species mappings of sensory neuron subtypes.
