Understanding the transcriptome changes in HeLa cells after inactivation of endosomes/lysosomes

ABSTRACT: In this study, we optimized an experimental approach to inactivate endo/lysosomal compartment, inducing the formation of new endosomes. Endo/lysosomes inactivation was executed through DAB- mediated depletion of endosomes/lysosomes in HeLa cells, which previously internalized Horseradish peroxidase (HRP). These cells, 18 hours after inactivation, can regenerate new endosomes capable of HRP uptake. We further observed the transcriptional response in HeLa cells driven by endo/lysosomes inactivation, discovering that, unlike the control condition, in which HRP was internalized but DAB solution was not added to the cells, a substantial number of endosomes/lysosomes related genes are activated
