RNA-seq analysis of skeletal muscle from control (Cre) and Terf2 knockout (KO) mice in gastrocnemius-plantaris-soleus (GPS) and tibialis anterior (TA)

ABSTRACT: To investigate the molecular effects of telomere damage in skeletal muscle by Terf2 knockout, we performed bulk RNA sequencing on gastrocnemius-plantaris-soleus (GPS) and tibialis anterior (TA) muscles isolated from Terf2 knockout (KO) and control (Cre) mice. Total RNA was extracted from dissected muscles and used for library preparation followed by paired-end sequencing. Samples include both male and female 8 month old mice. The dataset comprises biological replicates from KO and Cre genotypes for each muscle type. FASTQ files correspond to quality-filtered and trimmed reads used for alignment.

Project description:Global gene expression patterns were determined from microarray results on day 1, 3, 5, 7, 10 and 14 during plantaris muscle hypertrophy induced by synergist ablation in young adult mice (5 months). For each time point, two Affymetrix chips (Mouse Gene 1.0 ST) were used with 100 ng of total RNA derived from a pooled sample of either the right or left plantaris muscles from six animals.

Project description:Skeletal muscle is composed of multinucleated myofibers, in which nuclei share the cytoplasm, and other cell types. Here, we performed single-nucleus RNA sequencing (snRNA-Seq) to determine the gene expressions in each nucleus in gastrocnemius and plantaris muscles collected from 12-week-old C57BL/6 wild-type mice. Our datasets also provided a basis for comparing gene expressions between myofibers and other cell types and discovering the novel cell populations in muscle tissues.

Overloading MuSCs

Project description:Skeletal muscle possesses the ability to adapt its size in response to milieus, which is called plasticity. Overload like resistance training induces the increment of muscle mass called muscle hypertrophy. Muscle stem cells (also known as muscle stem cells) function to supply new nuclei for myofiber during the overload in muscle. Using compensatory hypertrophy in plantaris muscles, we isolated MuSCs from plantaris muscles 4 days after surgery. Control MuSCs were also prepared from sham plantaris muscles.
