Spatial transcriptomic analysis of rat gut tissue reveals transcriptional changes associated with high-fat diet and tirzepatide treatment

ABSTRACT: This study investigates spatial gene expression changes in rat gut tissue under different dietary and treatment conditions using 10x Genomics Visium spatial transcriptomics. Four experimental groups were analyzed: control diet with PBS treatment (CD_PBS), control diet with tirzepatide treatment (CD_TZP), high-fat diet with PBS treatment (HFD_PBS), and high-fat diet with tirzepatide treatment (HFD_TZP). Spatial transcriptomic libraries were generated using the 10x Genomics Visium platform and sequenced on an Illumina sequencing platform. Raw sequencing data were processed using the SpaceRanger pipeline to generate gene-barcode matrices and spatial coordinates. Downstream analyses included cell type annotation, differential gene expression analysis, and gene ontology enrichment analysis. These data provide insight into the spatial molecular mechanisms associated with metabolic disease and therapeutic intervention.
