RNA-seq of human iPS cell-derived retinal ganglion cells

ABSTRACT: The integrin family is a key regulator of cellular behavior, transducing adhesion signals through interactions with the extracellular matrix (ECM). Despite its central role in retinal ganglion cell (RGC) survival and neurite outgrowth, the comprehensive expression profile of integrins in human RGCs remains poorly characterized. In this study, we performed RNA sequencing on human iPSC-derived RGCs to elucidate their global transcriptomic landscape. Following a modified Sluch et al. (2017) protocol, RGCs were differentiated from 201B7 iPSCs and isolated at day 40 via CD90.2/Thy1.2-targeted magnetic-activated cell sorting (MACS). Our analysis confirmed robust expression of canonical RGC markers and revealed a significant enrichment of genes involved in ECM-mediated signaling, providing new insights into the molecular basis of RGC-scaffold interactions.
