ABSTRACT: Postmenopausal osteoporosis (PMOP) is characterized by high-turnover bone loss and oxidative stress. Astragaloside IV (AS-IV) exhibits antioxidant and immunomodulatory activities, yet its skeletal effects and mechanistic basis in ovariectomized (OVX) models remain incompletely defined.

Nat10-mediated N4-acetylcytidine modification enhances Nfact1 translation to exacerbate osteoclastogenesis in postmenopausal osteoporosis [RNA-seq]

Project description:Increased differentiation or activity of osteoclasts is the key pathogenic factor of postmenopausal osteoporosis (PMOP). N4-acetylcytidine (ac4C) modification, catalyzed by Nat10, is a novel post-transcriptional mRNA modification related to many diseases. However, its impact on regulating osteoclast activation in PMOP remains uncertain. Here, we initially observed that Nat10-mediated ac4C positively correlates with osteoclast differentiation of monocytes and low bone mass in PMOP. The specific knockout of Nat10 in monocytes and remodelin, a Nat10 inhibitor, alleviates ovariectomized (OVX)-induced bone loss by downregulating osteoclast differentiation. Mechanistically, epitranscriptomic analyses reveal that the nuclear factor of activated T cells cytoplasmic 1 (Nfatc1) is the key downstream target of ac4C modification during osteoclast differentiation. Subsequently, translatomic results demonstrate that Nat10-mediated ac4C enhances the translation efficiency of Nfatc1, thereby inducing Nfatc1 expression and consequent osteoclast maturation. Cumulatively, these findings reveal the promotive role of Nat10 in osteoclast differentiation and PMOP from a novel field of RNA modifications and suggest that Nat10 can be a target of epigenetic therapy for preventing bone loss in PMOP.

Artemisiae Scopariae Herba (Yinchen, YC) extracts have been reported to promote osteogenic differentiation and bone formation, but the specific mechanism is still unclear. Aim of the study: To investigate the effect and mechanism of YC on PMOP. Methods: Ultra-performance liquid chromatography-tandem mass spectrometry was used to determine the main potential mechanism of YC, and an ovariectomized (OVX) mouse model was established to evaluate the effects of YC on PMOP and to explore its potential mechanisms. First, the therapeutic effect of YC on PMOP was assessed by CT bone analysis, pathological observation and ELISA detection. Combined with serum ELISA, qRT-PCR and immunohistochemical staining, the potential key anti-PMOP pathway of YC was further explored.</p>

The novel tRF-23 promotes osteogenic differentiation of hBMSCs and protects against bone loss in ovariectomized mice

Project description:The pathogenesis of osteoporosis is closely related to the impaired human bone marrow-derived stromal cells (hBMSCs) osteogenic differentiation. No studies to date, however, have established whether tRNA-derived fragments (tRFs) can influence osteogenic differentiation of hBMSCs or the onset of osteoporosis. Here, tRF-23 was found to control hBMSC osteogenesis through its ability to target suppressor of cytokine signaling 1 (SOCS1) via the Janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3) signaling pathway. tRF-23 was then further established as a potential target for efforts to protect against bone loss and marrow adipose tissue (MAT) accumulation in osteoporotic model mice, and its molecular mechanism was also verified in vivo. Together, these results suggest a model in which tRF-23 can protect against bone loss induced by ovariectomized (OVX) through the augmentation of hBMSC osteogenesis, providing a foundation for further characterizing the pathogenesis of osteoporosis and seeking new therapeutic targets for this disruptive condition.

Project description:In postmenopausal osteoporosis (PMOP), the differentiation of bone marrow mesenchymal stem cells (BMSCs) is unbalanced,because of the lack estrogen, that is, osteogenic differentiation is reduced, adipogenic differentiation is increased.To investigate the mechanism of aberrant differentiation of BMSCs in PMOP, we performed transcriptome sequencing of Bone marrow mesenchymal stem cells in a postmenopausal osteoporosis model(PMOP-BMSCs)and Bone marrow mesenchymal stem cells in a healthy control model(Normal-BMSCs).In this analysis, the reference transcriptome sequencing of 6 samples was completed, and the genome alignment of each sample was obtained by aligning the reads to the reference genome, and the expression of protein-coding genes was analyzed based on the alignment results. According to the expression of protein-coding genes in different samples, differential screening was performed, and a total of 1 differential group was set up.
