CRISPRi lncRNA depletion screen in acute myeloid leukemia cell lines

ABSTRACT: To identify long non-coding RNAs (lncRNAs) essential for proliferation and survival of acute myeloid leukemia (AML) cells, we performed a genome-wide CRISPRi depletion screen using a custom CRISPRi lncRNA library. Six AML cell lines (K562, THP-1, ML-2, SKNO-1, M07e, NOMO-1) were screened with two biological replicates each. sgRNA representation was measured at baseline (Day 0) and after 18 population doublings. Screen hits were identified using the MAGeCK MLE algorithm with copy number variation correction.
