Immune niche analysis in breast tumour derived lung metastasis

ABSTRACT: We used our Cherry-niche labelling system to specifically isolate immune cells from the metastatic niche of established 4T1 lung metastases and compared their profile with immune cells isolated from the distal lung away from the tumour growth.

Project description:This experiment looks at the dissection of the microenvironment in the lung metastatic niche in a model of murine triple-negative breast cancer as disease progresses. Mice received an orthotopic inoculation of 4T1 cells and disease was allowed to progress for 7, 14, or 21 days correlating to the pre-metastatic, micro-metastatic, and metastatic niche, respectively. Healthy controls were obtained along with each time point.

Project description:Expression data from 4T1 subclones derived from mammary fat pad tumors (MFP), axillary lymph node tumors (AxLN), and axillary lymph node-derived lung metastases (AxLN-LuM). In parallel, expression data, in the same subclones, of tail vein-derived (TV) lung metastases. The mechanism of how lymph node metastases seed distant metastases is unknown. We used the 4T1 breast cancer cell line, which is an immune competent model of triple negative breast cancer and spontaneously metastasizes in balb/c mice. 4T1-GFP/fLuc cells were injected into MFP to form tumors and 4T1-mCherry/rLuc cells were injected into axillary lymph nodes to form tumors and then allowed to metastasize to lung. TV cells were allowed to metastasize in the lung. Cells were harvested at different time intervals after the injection. Tumors were extracted, dissociated, and then expanded in vitro to obtain MFP, AxLN, AxLN-LuM and TV-LuM subclones isolated after different time lags with respect to the injection.

Single cell RNA-seq of an native and engineered metastatic niche in a metastatic breast cancer murine model

Project description:Metastatic lesions are typically not found until patients self-report symptoms or they become radiologically evident. We have developed an engineered metastatic niche (scaffold) that recruits aggressive tumor cells prior to their colonization in other organs. The engineered niche can be monitored for dynamic gene expression, and changes at this site are analogous to those in a native metastatic site (lung) for triple negative breast cancer (4T1 cells). We were able to develop a 10-gene signature from the scaffold that accurately monitors disease progression and recurrence or resistance to resection therapy. This data set acts to dissect the heterogeneity of the cell populations in the engineered and native metastatic niche and identify the cell types that contribute to the success of the signature.

S100A4 inhibition alters pre-metastatic niche formation in 4T1 lung metastasis model

Project description:Cancer metastasis poses a major clinical challenge and is responsible for ~90% of cancer-associated deaths. However, progress in treating metastatic disease is limited. S100A4/FSP1 has long been associated with cancer metastasis, including breast cancer. In this study, we report preclinical efficacy data of a novel S100A4 blocking antibody that suppresses breast cancer metastasis. 4T1 and MMTV-PyMT breast cancer metastasis to the lung is greatly attenuated by S100A4 blocking antibody treatment. Immunohistochemistry analysis shows that neutrophil infiltration is significantly reduced in the anti-S100A4 treated mice. Single-cell RNA sequence analysis of the premetastatic niche treated with either isotype control or anti-S100A4 antibody from the 4T1 model show that NK and T cell activation in the lung. In this study, we report that S100A4 promotes premetastatic niche formation by promoting neutrophil infiltration and suppressing the function of cytotoxic NK and T cells.

A novel labelling strategy enables spatial resolution of metastatic niche cellular composition and uncovers lung stem cell activity proximal to breast cancer

Project description:Cancer cell behaviour is strongly influenced by the surrounding cellular environment, making the characterization of the local tumour microenvironment (or niche) a fundamental question in tumour biology. To date, a direct investigation of the early cellular changes induced by metastatic cells within the surrounding tissue is difficult to achieve, especially at early micro-metastatic stages and for low frequency niche populations. Here we present the strategy whereby metastatic cancer cells release a cell-penetrating fluorescent protein that is efficiently taken up by neighbouring cells, allowing spatial identification of the local metastatic cellular environment within the whole tissue. Notably, this strategy can be used to follow metastatic niches from early micro-metastasis to late macro-metastasis, allowing temporal resolution. Moreover, the presence of low represented niche cells can be detected and characterized among the bulk tissue. To highlight its potential, we have used this niche-labelling strategy to study the lung metastatic environment of breast cancer cells. We uncover the presence of lung parenchymal cells within the metastatic niche where lung epithelial cells show stem cell-like features with expression of lung progenitor markers, multi-lineage differentiation potential and self-renewal activity. Moreover, lung epithelial cells can be directly perturbed by cancer cells in ex vivo co-culture assays and support their growth. In summary, here we describe a novel labelling system that enables spatial resolution of the metastatic microenvironment and provide evidence that the tissue cellular environment surrounding metastatic growth is characterized by undifferentiated features. The data highlight the significant potential of this method as a platform for new discoveries.

Subtherapeutic-Dose of Ionizing Radiation Reprograms the Pre-Metastatic Lung Niche, Accelerating Its Formation and Promoting Metastasis

Project description:Pre-metastatic niche (PMN) formation is a critical step in metastatic progression, yet the biological effect of subtherapeutic-dose of ionizing radiation (SDIR) following radiotherapy on this process remains unclear. Using a 4T1 breast cancer mouse model, we investigated the effects of SDIR (3×0.3 Gy) on lung PMN development and metastasis formation. Lung PMNs were exposed to SDIR on days 8–10 post-tumor induction, followed by mastectomy on day 18, and analyzed on day 24. SDIR significantly increased total metastatic volume (TMV) in lungs, suggesting an accelerated PMN formation. Mechanistically, SDIR upregulated Bv8 expression, enhanced neutrophil recruitment, and increased MMP9, S100A8, and IL-6 production in the PMN by day 11, though differences between irradiated and non-irradiated PMN were no longer evident by day 14. Notably, Bv8 inhibition prevented SDIR-induced neutrophil recruitment but did not reduce TMV, indicating additional mechanisms in SDIR-driven metastasis. Proteomic analysis revealed SDIR-induced metabolic reprogramming, extracellular matrix (ECM) remodeling, and upregulation of adhesion-related pathways, driving a microenvironment that accelerates tumor invasion and metastatic outgrowth. By demonstrating that SDIR reprograms the pre-metastatic lung microenvironment, this study highlights the need to integrate organ-specific radiation exposure into predictive models of metastasis and identifies metabolic and immune-stromal pathways as potential targets for therapeutic intervention.
