Defined cell culture medium contributes to transcriptional reprogramming to enhance membrane-associated mucin expression in human corneal epithelial cells

ABSTRACT: This study investigates the transcriptional mechanisms by which a defined culture condition enhances membrane-associated mucin expression in human corneal epithelial cells (HCECs). RNA sequencing was performed on HCECs cultured in keratinocyte serum-free medium (KSFM) or a defined special medium. Transcriptomic analysis revealed coordinated activation of epithelial maintenance and mucin biosynthesis pathways. These findings provide insight into epithelial homeostasis and suggest potential therapeutic strategies for dry eye disease.
