Effect of Rv2282c, Rv1990c and Rv1985c overexpression on gene expression under ATc induction in Mycobacterium smegmatis [batch 1]

ABSTRACT: transcriptional regulation on DNA damage response is essential for persistence and mutagenesis of Mtb. We here report Rv2282c, Rv1990c and Rv1985c as regulators for DNA damage response system. Overexpression of Rv2282c, Rv1990c and Rv1985c greatly induce DNA replication and repair in a LexA-independent pathway, and inhibit cell division. Rv2282c, Rv1990c and Rv1985c induce DNA damage response by affecting redox state in mycobacterium smegmatis.Complemention of tpx relieves the effect of Rv2282c, Rv1990c and Rv1985c. Taken together, our study provides a conceptual framework for understanding how mycobacteria orchestrate the complex DDR system through redox-based transcriptional regulation.
