Single-nucleus transcriptomic atlas of the mouse medial thalamus


ABSTRACT: The mediodorsal thalamus and adjacent midline nuclei are critical for higher-order cognition and neuropsychiatric disease, yet their cellular organization remains incompletely defined. The objective of this study was to characterize the cellular composition and transcriptional organization of the mouse medial thalamus. Single-nucleus RNA sequencing was used to identify neuronal cell types, assign transcriptional clusters to anatomical nuclei, and define gene expression patterns underlying thalamic organization. A further goal was to compare these features across species to identify conserved transcriptional patterns and differences in cell type composition, including neuronal populations expanded in humans relative to mice. This dataset provides a transcriptomic atlas of the medial thalamus and a resource for investigating cell type diversity, functional organization, and cross-species specialization.
