ScRNA-seq of cardiac neural crest cells following left atrial ligation

ABSTRACT: We have employed a single cell sequencing approach using 10x Genomics scRNAseq to study the affect of altered hemodynamic patterning on cardac neural crest cells (CNCCs). CNCCs play an important role in tunica media formation of the great vessels, cardiac septation, and contribute to cardiomyocytes of he ventricle.

Single-cell transcriptomics uncovers a non-autonomous Tbx1-dependent genetic program controlling cardiac neural crest cell development

Project description:Disruption of cardiac neural crest cells (CNCCs) results in congenital heart disease, yet we do not understand the cell fate dynamics as these cells differentiate to vascular smooth muscle cells. Here we performed single-cell RNA-sequencing of NCCs from the pharyngeal apparatus with the heart in control mouse embryos and when Tbx1, the gene for 22q11.2 deletion syndrome, is inactivated. We uncover three dynamic transitions of pharyngeal NCCs expressing Tbx2 and Tbx3 through differentiated CNCCs expressing cardiac transcription factors with smooth muscle genes. These transitions are altered non-autonomously by loss of Tbx1. Further, inactivation of Tbx2 and Tbx3 in early CNCCs results in aortic arch branching defects due to failed smooth muscle differentiation. Loss of Tbx1 interrupts mesoderm to CNCC cell-cell communication with upregulation and premature activation of BMP signaling and reduced MAPK signaling, as well as alteration of other signaling, and failed dynamic transitions of CNCCs leading to disruption of aortic arch artery formation and cardiac outflow tract septation.

Gene expression analysis of WT and Adam19KO cardiac neural crest cells

Project description:Neural crest cells (NCCs) migrate into the heart, and their defects lead to congenital heart diseases. Despite the importance of cardiac NCCs (CNCCs), their final fates and underlying mechanisms of the fate specification have been unknown. Here we report that CNCCs differentiate from bipotent progenitors to tenocytes by preventing chondrogenesis. CNCCs highly express a disintegrin and metalloprotease 19 (Adam19) after migrating into the heart. Adam19-deleted CNCCs fail to suppress expression of Sox9 and generate abnormal cartilage, while they normally form the tendon connecting the aorta and pulmonary artery. Highly expressed Sox9 in CNCCs is necessary and sufficient for their chondrogenic fate decision. Adam19 inhibits response to BMP, preventing CNCCs from enhanced Sox9 expression. Our study reveals a novel fate of the NCC, and moreover, provides insights into how postmigratory NCCs define their fates locally.

Left Atrial Appendage Ligation And Exclusion Technology In The Incubator.

Project description:Stroke is the most feared complication of atrial fibrillation (AF). Targeting the left atrial appendage (LAA) mechanically is attractive as a means to simultaneously reduce stroke risk, the need for anticoagulation, and hemorrhagic complications in patients with non-valvular AF. The results of the PROTECT-AF and PREVAIL randomized clinical trials support this approach as a viable therapeutic alternative to warfarin in selected patients and add to accumulating evidence regarding the importance of the LAA in thromboembolism in AF. A number of devices for percutaneous LAA closure are under investigation or development. In this article, key design features of these ligation and exclusion technologies will be discussed, with a focus on aspects of LAA morphology, relational anatomy, thrombosis, and thromboembolism relevant for successful device development and deployment.

Project description:Here we derive human and chimpanzee cranial neural crest cells (CNCCs) and profile histone modifications, transcription factors, chromatin accessibility and gene expression to systematically and quantitatively annotate evolutionary divergence of craniofacial cis-regulatory landscapes. Histone modifications (H3K27ac, H3K4me1, H3K4me3, H3K27me3), chromatin modifiers (p300), transcription factors (NR2F1, TFAP2A), chromatin accessibility (ATACseq) and gene expression (RNAseq) were assayed in CNCCs derived from iPSCs/ESCs from 2 chimpanzee and 3 human individuals.

Pulsatile stretch stimulates endothelial PDGFB secretion to maintain arterial vascular smooth muscle

Project description:The endothelial cells (ECs) that line blood vessels are remarkably sensitive to mechanical force. Hemodynamic force impacts ECs in three dimensions: laminar shear stress, intraluminal pressure, and circumferential stretch. The magnitude of these forces in arteries is pulsatile in nature—a quality that is lost in veins. The role of pulsatility in shaping these vessels is not well understood. Here we examine loss of pulmonary arterial pulsatility following the Glenn surgery (surgical palliation of single ventricle congenital heart disease). Using cultured pulmonary arterial ECs, we define the transcriptional changes driven by pulsatility within each dimension. We identify pulsatile stretch as a critical stimulus for endothelial secretion of PDGFB—a known driver of vascular smooth muscle cell (VSMC) proliferation—and show that loss of arterial pulsatility in vivo leads to thinning of VSMCs. This work provides a mechanistic understanding of how arterial pulsatility maintains the structure that supports arterial hemodynamic force.

Mechanisms of TGFB1 induced atrial fibrosis

Project description:This project aims to compare gene expression in transgenic mice that overexpress TGFB1 to wild type mice. Transgenic mice have a fibrotic response in the atria, but not the ventricle. This is a 2 color design; dye swaps and a loop design were incorporated. Each group has N=4, with the exception being WT atria which has N=3 and was hybridied twice, the groups are Wt atria, WT ventricle, Transgenic atria, Transgenic ventricle.
