Single-cell transcriptomic analysis of SP11-treated MOLT4 cell line and untreated control.

ABSTRACT: We used single-cell RNA sequencing with the 10x Genomics platform to study the effect of SP11 treatment in a MOLT4. The goal was to examine tumor heterogeneity, identify potential treatment-resistant cell populations, and assess changes in the intercellular communication. This dataset provides an overview of how SP11 impacts tumor biology at single-cell resolution.

Project description:Here, we present global microprotein quantitation in two human leukemia cell lines, K562 and MOLT4. We identify unannotated microproteins that are differentially expressed in these cell lines. The expression of six microproteins was validated biochemically, and two were found to localize to the nucleus. Thus, we demonstrate that label-free comparative proteomics enables quantitation of microprotein expression between human cell lines, and that differentially expressed microproteins have properties, like subcellular localization, consistent with functionality.

Project description:MicroRNAs can play important roles in gene regulation affecting both normal development and diseases, including cancer. This microarray experiment was performed to identify genes and pathways differentially expressed in MOLT4 T-leukemia cells engineered to overexpress hsa-pre-miR-22-3p. MOLT4 cells transduced with an empty or hsa-pre-miR-22-3p lentiviral vectors were collected, processed for RNA extraction and hybridized on Affymetrix Clariom™ S Human arrays (n=3 replicates/group). Raw microarray data are available together with the applied protocols.

scRNA-seq of immune cells during targeted therapy-induced tumor regression and targeted therapy tolerant residual melanoma

Project description:Single-cell RNA sequencing (scRNA-seq) was performed to profile immune cell states in BRAF-mutant melanoma during targeted therapy-induced tumor regression and therapy-tolerant residual disease. CD45⁺ tumor-infiltrating immune cells were isolated from immunocompetent melanoma models treated with BRAF/MEK inhibitors and analyzed by scRNA-seq using the 10x Genomics Chromium platform. Comparative single-cell transcriptomic analysis revealed dynamic remodeling of the tumor immune microenvironment during treatment, including distinct macrophage and NK cell populations associated with therapeutic response and tolerance. Macrophage–NK cell crosstalk and cytokine signaling pathways were found to orchestrate immune activation during tumor regression, whereas immunosuppressive macrophage subsets and reduced NK effector signatures characterized the tolerant state. This dataset provides a comprehensive immune cell–level resource to investigate innate immune dynamics and intercellular communication mechanisms underlying targeted therapy efficacy and resistance in melanoma.
