Project description:In maize crops Southern Leaf Blight (SLB) is a severe disease caused by the fungus Bipolaris maydis (Y. Nisik & C. Miyake) leading to several losses in crop production. Unlike in maize, popcorn resistant varieties are not yet commercially available and this trait have been the subject of many popcorn breeding programs. The present study aimed to identify differentially accumulated proteins (DAPs) associated with resistance to B. maydis in two contrasting popcorn inbred lines using the comparative proteomic analysis. Forty-day old popcorn plants from resistant and susceptible inbred lines at the V4 growth stage were inoculated with B. maydis by spraying the conidium suspension. At four and ten days after inoculation, the morphological aspects of lesions in leaves were checked and the proteomic analysis was performed. Our results showed that the resistant inbred line exhibited minor foliar lesions in comparison with the SLB-susceptible genotype. In the genotype’s comparison, 644 DAPs were identified at 4 days and 613 at 10 days after B. maydis inoculation. Besides that, in resistant plants were identified DAPs upaccumulated related to response to stress, response to stimulus, photosynthesis, cellular growth, maintenance and detoxification process, which might be involved in plant response against the pathogen. It was identified proteins as salicylic acid-related and nucleotide-binding leucine-rich repeat that may be responsible to initiate a response to fungus in SBL-resistant inbred line. A differential response was identified in chloroplast proteins that is the main organelle involved in perception of the fungus to initiate a signalizing response. The changes in the proteomic profile in resistant inbred line may be effective in the response under B. maydis infection. This is the first work presenting the proteomic profile alteration under B. maydis inoculation and these findings may be useful for identifying candidate biomarkers in popcorn resistance to B. maydis being relevant for further researches on genetic breeding to develop resistant genotypes.

Project description:Exserohilum turcicum is a fungal pathogen that causes northern corn leaf blight and sorghum leaf blight. However, strains are host-specific, meaning a strain typically only causes disease on either maize or sorghum. This pathosystem provides a unique opportunity to identify the mechanisms underlying host specificity through genomic and transcriptomic studies. To characterize host specificity, we (i) constructed de novo annotated assemblies of one sorghum- (15St008) and one maize-specific (Et52B) E. turcicum strain, ii) identified and characterized structural changes between the two genomes, and (iii) compared differential gene expression. This GEO project contains the gene expression of the maize-specific strain (Et52B) during in planta infection of maize and in axenic cultures.

Project description:Tar spot of maize, caused by Phyllachora maydis, is an emerging threat to crop production across the United States and Canada. Current effective management of the disease includes application of fungicides and use of partially resistant maize varieties. Several studies have focused on mapping of tar spot resistant loci from maize diversity panels. However, no additional analyses have been reported to further our understanding of the maize defense response to P. maydis. Therefore, prior to the availability of inoculation procedures, we performed transcriptome sequencing from maize leaf tissue exposed to P. maydis from a tar spot infested field and assessed differential gene transcript expression. Over the course of disease development, leaves were sampled at two timepoints: 10 days and 24 days post-exposure. Differentially expressed genes (DEGs) were determined by comparing gene transcript expression of the exposed samples to non-exposed controls at the respective timepoints.

Project description:Plant pathogens secrete effectors, which target host proteins to facilitate infection. The Ustilago maydis effector UmSee1 is required for tumor formation in the leaf during infection of maize. UmSee1 interacts with maize SGT1 and blocks its phosphorylation in-vivo. In the absence of UmSee1, U. maydis cannot trigger tumor formation in the bundle sheath. However, it remains unclear which host processes are manipulated by UmSee1 and the UmSee1-SGT1 interaction to cause the observed phenotype. Proximity-dependent protein labeling involving the turbo biotin ligase tag (TurboID) for proximal labeling of proteins is a powerful tool for identifying the protein interactome. We have generated transgenic U. maydis that secretes biotin ligase-fused See1 effector (UmSee1-TurboID-3HA) directly into maize cells. This approach, in combination with conventional co-immunoprecipitation allowed to identify additional UmSee1 interactors in maize cells. Collectively, our data identified three ubiquitin-proteasome pathway-related proteins (ZmSIP1, ZmSIP2, ZmSIP3) that either interact with or are close to UmSee1 during host infection of maize with U. maydis. ZmSIP3 represents a cell cycle regulator which degradation appears to be promoted in the presence of UmSee1. Our data provide a possible explanation for the requirement of UmSee1 in tumor formation during U. maydis - Zea mays interaction.

The interaction of U. maydis with maize is a well-established model to study the interaction between maize and biotrophic pathogen. U. maydis infection could activate host immune responses including: ROS accumulation, protease activation, salicylic acid signaling. U. maydis employ several strategies to overcome maize immune response, thus initial the biotrophic interaction with host. It has been suggested that genetic factors of maize host affected the disease severity of U. maydis infection, here we investigated the transcriptome profile of resistance and susceptible maize lines upon U. maydis infection, thus propose candidate maize genes involved in the defense response in maize to corn smut cause by U. maydis.

Upon infection, Sts2 is translocated into maize cell nucleus, where it acts as a transcriptional activator, and the transactivation activity is crucial for its virulence function. Sts2 interacts with ZmNECAP1, a yet undescribed plant transcriptional activator, and it activates the expression of several leaf developmental regulators to potentiate tumor formation. Contrary, a suppressive Sts2-SRDX inhibits the tumor formation by SG200 in a dominant negative way, underpinning the central role of Sts2 for tumorigenesis. Our results not only disclosed the virulence mechanism of a tumorigenic effector, but also revealed the essential role of leaf developmental regulators in pathogen-induced tumor formation.
