Transcriptomic responses following AMC-EVs treatment in subfertile mares


ABSTRACT: Equine endometrial dysfunction is a major cause of subfertility in mares, and although the Kenney and Doig classification is widely used for prognosis, it does not capture the underlying molecular alterations. In this study, the early transcriptomic effects of intrauterine administration of amniotic membrane-derived extracellular vesicles (AMC-EVs) were investigated in subfertile mares using a paired pre- and post-treatment design. RNA sequencing revealed significant gene expression changes (FDR < 0.05) following treatment, including upregulation of pathways related to receptor-mediated signaling, immune response, and vesicular transport, alongside downregulation of metabolic and RNA-processing pathways. These findings indicate that AMC-EVs induce early transcriptional reprogramming in the equine endometrium, suggesting a potential role in modulating molecular processes associated with endometrial function.
