Effect of Laminin N terminus alpha31 on pancreatic adenocarcinoma formation

ABSTRACT: In this study we induced expression of the laminin alpha 3 splice isoform LaNt a31 in MiaPaca-2 pancreatic adenocarcinoma cells and implanted those cells into chicken egg chorioallanotoic membrane (CAM). Total mRNA seq was performed to identify genes and pathways changed through expression of this protein.

Gene expression of Foxp3+CD8+ Tregs induced with or without laminin α5

Project description:Foxp3+CD8+ regulatory T cells (CD8 Tregs) help to establish immune tolerance in models of chronic inflammation and antigen stimulation, such as graft versus host disease and chronic infections. However, the elements contributing to CD8 Treg induction remain obscure. In this study, we found that laminin α5 enhanced CD8 Treg inducation from mouse naive T cells stimulated with a combination of anit-CD3/anti-CD28/IL-2/TGFβ. While, the suppressive ability of CD8 Tregs induced with laminin α5 was reduced by 23.8% compared to these induced without laminin α5; the cytotoxic ability of CD8 Tregs induced with laminin α5 was significant reduced (more than 64%) compared to these induced without laminin α5. Mechanistically, laminin α5 was invovled in modulating both TCR signaling and cytokine responses during induction of CD8 Tregs. Laminin α5 inihbited IFNg and TNFa expression by CD8 T cells, which further enhanced Foxp3+CD8+ Treg induction. Laminin α5-Itga6 enhanced TCR signaling by promoting activation of the LAT/Zap70/PLCγ1 pathways.

Transcriptome analysis reveals differentially expressed genes associated with high rates of egg production in chicken hypothalamic-pituitary-ovarian axis

Project description:Purpose: To identify the key regulatory genes and pathways involved in chicken high egg productivon in HPG axis. Methods: A total of 856 Chinese Luhua chicken was raised in poultry breeding farm of Sichuan Agricultural University, the highest two hundred and the lowest two hundred chicken egg production were considered as high egg production (HEP) and low egg production (LEP) according to the total egg number at 300 days of age, respectively, integrated with RNA-seq sequencing of samples of HPG axis (hypothalamus, pituitary gland and ovary) from three HEP and three LEP chickens at 300 days of age. Results: A total 86.7 Gb RNA-seq sequences were generated, and with each library averaged 5.1 Gb. Conclusions: These important data might improve our understanding of reproductive biology of Luhua chicken by providing comprehensive gene expression information at transcriptional level. We indicate that our approach will contribute to the isolation of effective molecular markers that can be used in genetic breeding programs in Chinese domestic Luhua chicken.
