Project description:In present study, it was found that the co-culture of Wickerhamomyces anomalus Y-5 and L. plantarum RX-8 could enhance bacteriocin production. To analyze the interaction between W. anomalus Y-5 and L. plantarum RX-8, a quantitative proteomic approach was used to analyze and compare the proteome in L. plantarum RX-8 and W. anomalus Y-5 under mono-culture and co-culture. In total, 339 differently expressed proteins (DEPs) were screened in comparison of L. plantarum RX-8 under mono-culture and co-culture, 645 proteins of W. anomalus Y-5 changed in mono-culture and co-culture. The Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis indicated that DEPs participated in various metabolic pathways such as PTS system, glycolysis, galactose metabolism, glutamate, aspartate, arginine and cysteine metabolism etc. These pathways were related to inducing mechanism on improving bacteriocin production by co-culture. Quantitative proteomic analysis-based strategies can therefore provide further evidence for new regulated targets to improve the production of bacteriocins.

Project description:Probiotics have been suggested to ameliorate the function of the intestinal epithelial barrier and so have several mediators and receptors of the expanded endocannabinoid system, the endocannabinoidome (eCBome). Here we cocultured three live strains of Lactiplantibacillus plantarum with intestinal epithelial organoids to study their effects on the gut barrier function and the possible involvement of the eCBome in this effect. All three L.plantarum strains variously reduced the trans-epithelial permeability of intestinal organoids and promoted increased mRNA expression of several tight junction proteins and intestinal barrier proteins. Concomitantly, the three strains upregulated the expression of genes encoding biosynthetic enzymes (i.e., NapePLD, Abdh4, Gde1, Daglb) and receptors (i.e., Cnr1, Cnr2, Gpr55, and Ppara), while concurrently downregulating the expression of two essential catabolic enzymes (i.e. Faah and Naaa), involved in the signaling of several eCBome mediators known for their role in regulating the intestinal epithelial barrier. Selective inhibitors of eCBome mediator degrading enzymes FAAH and MAGL, i.e., URB597 and JZL184, increased N-acyl-ethanolamine (NAE) and 2-monoacylglycerol (2-MAG) levels, respectively, enhanced the expression of intestinal epithelial barrier genes and reduced the trans-epithelial permeability of organoids, as for L. plantarum strains. Interestingly, inflammation-induced trans-epithelial permeability in organoids was also reversed by both FAAH and MAGL inhibitors. We surmise that elevated endogenous levels of either NAEs or 2-MAGs promote improvement in small intestine trans-epithelial permeability and that L. plantarum strains may exploit this mechanism to promote these beneficial effects.

Project description:Light is a key environmental factor in the growth, development, and metabolism of the edιble mushroom Pleurotus ostreatus. In this study, the effect of white, blue, green, yellow, red light, and darkness on the global protein expression profile of P. ostreatus LGAM 1123 grown in submerged cultivation was investigated. No inhibition of the growth of the fungus, in all the light conditions tested, compared to the dark, was observed. However, the mycelial protein content was reduced by 10% in blue and white light. From the proteomic analysis, a different proteome was expressed for each light wavelength, with red and blue light presenting the most distinctive proteome profiles. Blue light induces pathways such as the citrate cycle (TCA cycle), glycolysis/gluconeogenesis and biosynthesis of amino acids, while red light induces mRNA-related pathways. GC-MS analysis of biomass revealed differences in the produced amino acids, sugars, and lipids. The distinct regulation of proteins and bioactive compounds under different light wavelengths growth indicate that using a specific light wavelength, the metabolism of P. ostreatus could turn into particular biochemical pathways. These strategies could be useful for the food industry because specific nutrients could be increased in the fermentation of edible fungi without the need for genetic engineering of the strain.

The effect of light on genome-wide transcriptional regulation has been well-studied in green algae and plants, but not in red algae. Cyanidioschyzon merolae is used as a model red algae, and is suitable for studies on transcriptomics because of its compact genome with a relatively small number of genes. In addition, complete genome sequences of the nucleus, mitochondrion, and chloroplast of this organism have been determined. Together, these attributes make C. merolae an ideal model organism to study the response to light stimuli at the transcriptional and the systems biology levels. Previous studies have shown that light significantly affects cell signaling in this organism, but there are no reports on its blue light- and red light-mediated transcriptional responses. We investigated the direct effects of blue and red light at the transcriptional level using RNA-seq. Blue and red light were found to regulate 35% of the total genes in C. merolae. Blue light affected the transcription of genes involved protein synthesis while red light specifically regulated the transcription of genes involved in photosynthesis and DNA repair. Blue or red light regulated genes involved in carbon metabolism and pigment biosynthesis. Overall, our data showed that red and blue light regulate the majority of the cellular, cell division, and repair processes in C. merolae.

Project description:Light quality is an important abiotic factor that affects growth and development of photosynthetic organism. In this study, D. salina was exposed to red (660 nm) and blue light (450 nm), and cell growth, pigments, and transcriptome were analyzed. The RNA of D. salina was sequenced and transcriptomic response of algal cells after transitioning from white light to red and blue light was investigated. Genes encoding for enzymes involved in photosynthesis were down-regulated, whereas genes involved in the metabolism of carotenoid were up-regulated. Genes encoding for photoprotective enzymes related to reactive oxygen species scavenging were up-regulated under both red and blue light. The present transcriptomic study would assist in the comprehensive understanding of carotenoid biosynthesis of D. salina.
