RNA-seq of cortical CD51+ astrocytes expressing hM3D-mCherry at P21 after saline or cno injection from P7 to P21

ABSTRACT: We injected transgenic CD51-Flp mice with AAV-GFAP-FSF-hM3D-mCherry postnatally. Mice were injected with saline or CNO from P7 to P21 twice per day. mCherry+ astrocytes were isolated from cortex at P21 by FACS for RNA-seq.

Gene Expression after Cochlear Removal in Cochlear Nucleus at P7 and P21

Project description:Deprivation of peripheral nerve input by cochlear removal in young mice results in dramatic neuron death in the cochlear nucleus (CN). The same manipulation in older mice does not result in significant loss. The molecular basis of this critical period of vulnerability remains largely unknown. Here we identified genes regulated at early time points after cochlear removal at ages when neurons are vulnerable (postnatal day (P)7) or invulnerable (P21) to this challenge. Afferent deprivation regulated very different sets of genes at P7 and P21. These genes showed a variety of functions at both ages, but surprisingly there was no net increase in pro-apoptotic genes at P7. A large set of upregulated immune-related genes was identified at P21. Experiment Overall Design: Mice received unilateral cochlear removals. At 6, 12, 24, and 48 hours after surgery, the CN ipsilateral and contralateral were removed, and RNA isolated from separate pools of tissue for each replicate. Deafferented CN were compared to age-matched and time-matched contralateral, control CN to identify genes regulated by cochlear removal at age P7 and P21.

CD51 expression delineates functionally distinct astrocyte subpopulations in neural circuits

Project description:Astrocytes execute a diverse range of tasks that facilitate brain physiology, raising the possibility that these diverse astrocytic roles are enabled by functionally distinct subtypes1,2. Recent studies identified molecular diversity amongst astrocytes3, however whether this molecular heterogeneity reflects specialized astrocyte subpopulations with distinct functions remains unknown. Leveraging prior molecular and functional studies4, we generated a CD51-FLP mouse line that enables the direct comparison of functional properties between CD51+ and CD51- astrocyte subpopulations in neural circuits in vivo. Here we show that CD51+ and CD51- astrocytes are endowed with unique morphological, physiological, and developmental properties in both the cortex and hippocampus. Functional studies demonstrate that CD51+ astrocytes selectively promote synapse formation in the developing cortex and circuit plasticity in the hippocampus. Mechanistically, we found that CD51+ astrocytes function through glypican 6 to promote synapse formation in the cortex, while CD51 itself drives circuit plasticity in the hippocampus. Applying these findings to brain injury, we found that exploiting the unique properties of CD51+ astrocytes promotes functional recovery after stroke through enhanced synaptic remodeling. Taken together, our studies identify specialized astrocyte subpopulations that exhibit distinct functional properties in the developing, mature, and injured brain.

Gene expression profilling of NEUROG2-mCherry lineage in cerebral organoids

Project description:To identify and purify NEUROG2-expressing cells and trace their short-term lineage, we engineered two NEUROG2-mCherry knock-in human embryonic stem cell (hESC) lines. Transcriptomic profiling of NEUROG2:mCherry knock-in hESC-derived cerebral organoids revealed an enrichment of neurogenic, oligodendrocyte precursor cell and extracellular matrix-associated gene transcripts in mCherry-high cells. Conversely, only neurogenic gene transcripts were enriched in mCherry-high cells from Neurog2:mCherry knock-in mouse cortices.
