Immune remodeling after thymectomy in patients with anti-AChR antibodies


ABSTRACT: We performed single-cell RNA sequencing (scRNA-seq) and T cell receptor (TCR) sequencing of peripheral blood mononuclear cells (PBMCs) from anti-acetylcholine receptor (AChR) antibody–positive patients before and after thymectomy. Analysis of immune cell composition revealed a reduction in naïve T cell populations and an increase in CD56^dim NK cells following thymectomy. Transcriptional profiling identified activation-associated gene expression in T cells and substantial gene expression changes in monocyte subsets, including reduced expression of antigen presentation–related genes. In addition, cell–cell communication analysis revealed changes in dominant signaling pathways, with differences observed in TNF-, resistin-, and IFNγ-related signaling between pre- and post-thymectomy samples. Together, these data provide a resource for understanding immune remodeling after thymectomy at single-cell resolution.
