Tamarixetin induces DNA damage response and activates p53 signaling in A549 non-small cell lung cancer cells


ABSTRACT: Tamarixetin (TAM), a naturally occurring O-methylated flavonol, exhibits antitumor activity in non-small cell lung cancer (NSCLC), but its transcriptomic effects and molecular mechanism remain incompletely defined. In this study, RNA sequencing was performed to characterize gene expression changes in A549 NSCLC cells after treatment with TAM. A549 cells were treated with vehicle control or 25 μM TAM for 24 h, followed by transcriptome profiling. Differential expression and pathway enrichment analyses revealed that TAM treatment induced substantial transcriptional reprogramming, with enrichment of p53 signaling, DNA damage response, DNA repair, and cell-cycle regulatory pathways. These transcriptomic findings support the mechanism that TAM activates DNA damage response signaling and p53-dependent antitumor pathways in NSCLC cells.
