Reduced-representation bisulfite sequencing (RRBS) of mouse kidney in kidney-specific Glut2 knockout (rGlut2 KO)


ABSTRACT: Glycosuria, whether genetically induced or triggered by SGLT2 inhibitors, activates compensatory glucose-producing pathways that limit glucose lowering in type 2 diabetes. To define these pathways, we studied renal Glut2 knockout mice, which progressively lose Slc5a2 (encoding SGLT2) expression yet maintain normoglycemia despite marked urinary glucose loss. To characterize DNA methylation changes associated with chronic glycosuria, reduced-representation bisulfite sequencing (RRBS), in which genomic DNA was digested with MspI to enrich for CpG-rich regions, was performed on kidney tissue from 12-week-old male rGlut2 KO and wild-type control mice (3 biological replicates per group). Libraries were prepared and sequenced by ZymoResearch (project ID: zr24946). Reads were quality-trimmed using Trim Galore and aligned to the mouse reference genome (mm10) using Bismark, and CpG methylation was called using MethylDackel.
