Feature-specific threat coding in lateral septum guides defensive action

Project description:The ability to rapidly detect and evaluate potential threats is essential for survival and requires the integration of sensory information, with internal state and prior experience. The lateral septum (LS), an inhibitory structure in the limbic forebrain, is thought to integrate these higher-order ‘cognitive’ signals to regulate defensive responses1,2. However, the cellular, circuit, and computational mechanisms fundamental to this process remain unknown. Here, we focus on the LS population expressing the type 2 CRH receptor (LSCrhr2), a neuronal subset shown to be critical for state-dependent behavioral changes and threat responsivity3–7. We use a combination of single-cell calcium imaging, molecular sequencing, and circuit dissection to reveal the spatial and functional organization of the cell-types involved, the computations they perform, and the information relayed by their upstream activators. We determined that LSCrhr2 population activity is required for cuedriven defensive actions by rapidly and dynamically encoding threat representations that predict defensive outcomes. We find that these threat representations are formed through the convergence of various signals differentially represented among distinct LSCrhr2 subclasses, which are defined by their molecular features, spatial location, and input architecture. Most importantly, their responses reflect specific afferents from the hippocampus and hypothalamus that preferentially impart cue- and action-related signals, respectively. These findings establish a multifeatured organizational principle that underlies how LS mediates motivated behaviors in response to environmental challenge.

Coordinated excitatory and inhibitory circuits governing unpredictable threat induced anxiety

Project description:Anxiety is elicited by excessive apprehension about unpredictable threats. However, the neural circuit governing unpredictable threat induced anxiety remains unclear. Here, we found ventral bed nucleus of the stria terminalis (vBNST) GABAergic neurons displayed selective activation to unpredictable threats by means ofthrough coordinated excitatory input from insular cortex (IC) glutamergic neurons and inhibitory input from lateral nucleus of the amygdala (CeL) somatostatin (SOM) neurons. Using activity-dependent neuronal tagging technology, we found that unpredictable threat responsive cells in vBNST drive freezing and anxiety via projections to ventral lateral periaqueductal grey (vlPAG) and median nucleus of the amygdala (CeM) respectively. Finally, we identified KCNQ3 plays an essential role in hyperactivity of vBNST GABAergic neurons and induced anxiety. These data identified a forward inhibitory circuit that determine the selective activation of vBNST in unpredictable threat and anxiety, and suggest that Kcnq3 KCNQ3 channel acts as a promising target in treatment of anxiety disorder following unpredictable stress.

Bidirectional regulation of social memory by two distinct types of GABAergic neurons in the lateral septum

Project description:Humans and animals memorize and recognize familiar conspecifics during social interactions, known associal memory. While the hippocampus is crucial for social memory, the dynamic regulation social memory, particularly the negative regulation, remains unclear. Here, we identified two molecularly distinct subpopulations of GABAergic neurons in the mouse lateral septum(LS) expressing somatostatin(SST) and calbindin1(CB), respectively, with complementary distribution patterns. Activation of SST+ neurons enhances social memory acquisition, whereas activation of CB+ neurons inhibits it. Optogenetic manipulation of SST+ neurons regulates theta rhythm oscillations, correlating with their impact on social memory. SST+-dLS neurons and CB+-vLS neurons preferentially receive stronger excitatory inputs from the dorsal (vCA1d) and ventral(vCA1v) parts of vCA1, respectively. The vCA1d→SST+-dLS→lateral hypothalamic area (LHA) circuit positively regulates social memory acquisition, whereas thevCA1v→CB+-vLS→anterior hypothalamic nucleus (AHN) pathway negatively regulates it. This study uncovers two parallel subcortical neural circuits underpinning the dynamic regulation of social memory.
